辛伐他汀对大鼠成骨细胞骨形成蛋白2及其信号传导蛋白Smad1和Smad5表达的影响

【摘要】 目的:研究辛伐他汀对成骨细胞骨形成蛋白2(BMP-2)及其信号传导蛋白Sma和Mad相关蛋白(Smad)1、5表达的影响,探讨该药物影响成骨的可能机制。方法:组织块培养法培养原代成骨细胞,传代纯化后用钙钴法染色鉴定。对照组S0无药物干预,实验组选用含不同浓度[10~8(S1组)、10~7(S2组)、5×10~7(S3组)、10~6(S4组)mol/L]辛伐他汀培养液对传代的成骨细胞进行72 h干预,采用反转录PCR(RT-PCR)测得BMP-2、Smad1、Smad 5 mRNA相对表达量,使用单因素方差分析和SNK-q检验进行统计学检验,以得出不同浓度药物对BMP-2及其信号传导蛋白的影响。结果:BMP-2、Smad1、Smad5 mRNA相对表达量均随药物浓度(0、10~8、10~7、5×10~7、10~6mol/L)增高而增加。三者在辛伐他汀浓度为10~6mol/L时表达量均最高。备组数据两两比较除Smad5表达量在对照组S0与S1组间,S3与S4组间差异无统计学意义(P>0.05)外,备实验组与对照组差异均有统计学意义(P<0.05)。结论:辛伐他汀能在一定剂量范围上调BMP-2的表达,可能与诱导BMP-2的信号转导分子Smadl、Smad5 mRNA的表达相关。

【Abstract】 Objective:To investigate the possible mechanism of simvastatin,a cardiovascular system agent,which has been found a promotive effects in bone formation by inducing the differentiation and proliferation of osteoblast,involving bone morphogenetic protein-2(BMP-2),and its signal transducer;sekclsky mothers against dpp 1 and 5(Smadl,and Smad5).Methods:A continuous tissue cultivation method was used to separate and cultivate primary osteoblasts from cranium of SD neonate rats.The relative mRNA expression of BMP-2,Smadl and Smad5 of osteoblasts were determined by reverse transcription PCR(RT-PCR) after being exposed to simvastatin[0(S0),10-8(S1),10-7(S2),5×10-7(S3),10-6(S4) mol/L]for 72 hours.Results:The relative expression of BMP-2,Smadl and Smad5 was significantly increased in a dose-dependent manner after being treated with simvastatin(10-8-10-6 mol/L).The 10-6mol/L simvastatin showed the best promotive effects on the expression of BMP-2,Smadl and Smad 5.Conclusion;The increased expression of mRNA of BMP-2,Smadl and Smad5 may be involved in the possible mechanism of bone formation induced by simvastatin.
