PTCH表达与临床乳腺癌患者预后相关性研究

【摘要】 目的考察临床乳腺癌病理组织中Hedgehog通路关键因子PTCH的表达与乳腺癌患者临床病理参数及预后的关系。方法通过免疫组织化学检测150例乳腺癌患者肿瘤组织的PTCH的表达;应用χ2检验分析PTCH表达与乳腺癌临床病理参数的相关性;采用时序检验绘制Kaplan-Meie生存曲线及Cox回归多变量分析PTCH表达与患者总生存期(OS)及无病生存期(DFS)的关系。结果 PTCH蛋白在乳腺癌组织中的阳性表达率为67.3%(101/150),PTCH阳性表达与患者肿瘤大小、淋巴结转移及组织分级相关(P<0.05);PTCH阳性表达患者的DFS和OS均显著短于PTCH阴性表达的患者(P<0.05)。结论 PTCH异常表达与乳腺癌患者的淋巴结转移及不良预后密切相关。

【Abstract】 Objective To evaluate the relationship of PTCH expression,one of the major components of Hedgehog signaling pathway,with the clinicopathological features and survival of breast cancer patients.Methods The expression of PTCH was measured by immunohistochemical staining in 150 cases of human breast cancer tissues.The association of PTCH protein expression with the clinicopathological features in breast cancer tissues was analyzed by Pearson Chi-square.The association of the expression of PTCH with overall survival(OS)and disease-free survival(DFS)in breast cancer patients was analyzed by Kaplan-Meier survival analysis.The association of clinicopathological data with DFS and OS in breast cancer was analyzed by multivariate Cox regression analysis.Results The positive expression rate of PTCH in 150 cases of human breast cancer tissues was 67.3%(101/150).The positive expression of PTCH was positively associated with tumor size,histological grade,and lymph node metastasis in breast cancer tissues(P <0.05).PTCH expression was significantly associated with shorter DFS and OS of breast cancer patients(P <0.05).Conclusion The abnormal expression of PTCH is correlated with lymph node metastasis and poor outcome of human breast cancer.
