解脲支原体感染对男性不育者精浆活性氧与细胞因子的影响
Influence of Seminal Plasma Reactive Oxygen Species and Cell Factors in Infertile Men with Ureaplasma Urealyticum Infection
【摘要】 目的 研究解脲支原体(Uu)感染对男性不育者精浆活性氧(ROS)、细胞因子及精子质量的影响,探讨其在男性不育中的作用机制。方法 选择83例Uu感染男性不育者为实验组(Uu+不育组),30例无Uu感染男性不育者(Uu-不育组)和30例正常男性已育者为对照组(正常生育组),分别测定精浆丙二醛(MDA)、超氧化物歧化酶(SOD)、IL-6,IL-10,IL-18和TNF-α水平并分析其相关性。结果 Uu~+不育组MDA含量为19.56±5.22 nmol/ml,IL-6含量为58.31±8.94pg/ml,IL-18含量为38.16±17.02 pg/ml和TNF-α含量为42.68±11.18 pg/ml,均明显高于Uu~-不育组和正常生育组(t=4.35~20.43,P值均<0.001),而IL-10含量为8.62±2.98 pg/ml和SOD活性为95.36±20.03μmol/L,均明显低于Uu~-不育组和正常生育组(t=3.67~23.74,P值均<0.001)。相关分析发现,Uu~+不育组MDA与TNF-α,IL-18呈正相关(r=0.61,0.55,P<0.001),而与SOD活力和IL-10呈负相关(r=-0.55,-0.53,P<0.001)。结论Uu感染导致精浆活性氧增加、细胞因子异常表达并影响精子质量,检测男性不育者精浆活性氧和细胞因子对男性不育的治疗具有重要意义。
【Abstract】 Objective To study the influence of seminal plasma reactive oxygen species(ROS),cell factors and sperm quality in infertile male with Uu infection and explore its action mechanism in male infertility.Methods Chose 83 cases of male infertility with Uu infection as the experimental group(Uu~+ infertility group),30 cases of male infertility without Uu infection(Uu~- infertility group) and 30 normal men with children as a control(Normal fertility group).Respectively,determinate the levels of seminal plasma malondialdehyde(MDA),superoxide dismutase(SOD),IL-6,IL-10,IL-18 and TNF-α,and analyzed its correlation.Results In Uu~+ infertility group,the levels of MDA(19.56 ± 5.22 nmol/ml),IL-6(58.31±8.94pg/ml),IL-18(38.16 ± 17.02 pg/ml) and TNF-α(42.68 ± 11.18 pg/ml) were obviously higher than those in the other two groups 0 = 4.35~20.43,P value<0.001),and the level of IL-10(8.62±2.98 pg/ml) and SOD(95.36 ± 20.03 μmol/L)was lower than those in the other two groups(t=3.67 ~23.74,P value<0.001).Correlation analysis found that MDA of Uu~+ infertility group was positively correlated with TNF-α,IL-18(r=0.61,0.55,P value<0.001),and negatively correlated with SOD and IL-10(r=-0.55,-0.53,P value<0.001).Conclusion The results suggested that Uu infection caused the level of reactive oxidative increasing,cytokines counterbalance disorder and affected sperm quality.So it is of great significance for the treatment to test the levels of ROS and cytokines in patients with male sterility.
