不同配比壳聚糖/聚磷酸钙复合支架应用于半月板的生物相容性研究

【摘要】 目的评价对比不同配比壳聚糖/聚磷酸钙复合支架的体外生物相容性,观察半月板纤维软骨细胞在不同配比支架材料上的生长情况,选出最佳配比CS/CPP生物材料用于半月板的治疗。方法利用ADA将CS与CPP混合液交联,制备不同配比的CS/CPP复合支架材料。采用扫描电镜检测半月板细胞在支架材料上的生长情况。采用MTT法检测不同配比CS/CPP支架材料浸提液的毒性,通过细胞接种的方法评价支架材料的生物相容性。结果扫描电镜下,CS/CPP复合支架材料均呈三维多孔结构;细胞在支架材料上粘性良好,分布均匀,生长良好,其中配比为3∶7CS/CPP复合支架材料上细胞粘性最好,黏附细胞数量最多。不同配比CS/CPP复合支架材料不同浓度浸提液的毒性均不高于1级,全部合格,配比为3∶7CS/CPP复合支架材料浸提液细胞相对增殖高、毒性小,细胞相容性最好,与其它配比相比,差异有统计学意义(<0.05)。结论不同配比壳聚糖/聚磷酸钙均具有良好的细胞相容性,其中配比为3∶7CS/CPP复合支架材料生物相容性最好,有望成为组织工程半月板的支架载体。

【Abstract】 Objective To explore the different ratio of chitosan/poly calcium phosphate composite scaffolds correlation of cells, and its applied to study the feasibility of meniscal tissue engineering scaffold material. Methods CS CPP ADA will use a mixture of crosslinking, prepare different ratios of CS/CPP composite scaffolds. Using the scaffold growth scanning electron microscopy meniscal cells. Detecting toxicity of different proportions CS/CPP scaffolds extract using MTT assay to evaluate the biocompatibility of scaffolds by cell vaccination method. Results Under the scanning electron microscopy(sem), CS/CPP composite scaffolds were three-dimensional porous structure; Cells on the scaffolds good stickiness,uniform distribution, grow well, the ratio of 3∶7 CS/CPP composite scaffold cells on the viscosity, the best adhesion cell count most. Different ratio of CS/CPP composite scaffold material toxicity of different concentration of leaching solution are higher than that of grade 1, all qualified, the ratio of 3∶7 CS/CPP composite scaffold extract small relative high proliferation, cell toxicity, cell compatibility is best, compared with the other proportion, the difference was statistically significant( <0.05). Conclusion Different ratio of chitosan/poly calcium phosphate has good cell compatibility, the ratio of 3∶7 CS/CPP composite scaffold material biocompatibility is best, is expected to become the carrier meniscus tissue engineering scaffolds.
