云南小粒种咖啡果皮粗提取物对人脐静脉内皮细胞抗氧化损伤的研究

【摘要】 云南小粒咖啡已成为云南省热带特色产业。云南小粒咖啡果皮在制作咖啡制品时常作为副产物丢弃,然而小粒咖啡果皮也可以作为花青素的来源。以人脐静脉内皮细胞(Human Umbilical Vein Endothelial Cell,HUVEC)进行培养,通过不同浓度(100、200、300、400 mg/L)的云南小粒咖啡果皮提取物处理后,再利用400μmol/L过氧化氢处理建立氧化损伤模型,对云南小粒种咖啡果皮粗提物的抗氧化损伤活性进行研究。结果表明,加入不同浓度的咖啡果皮粗提物对于受到损伤的人脐静脉内皮细胞有一定的保护和恢复作用;在对细胞氧化应激相关物质的分析测定中发现,相比氧化损伤组,400 mg/L浓度的咖啡果皮粗提物处理组提高了细胞中SOD活性(9.6±0.35)%,MDA含量降低了(23.0±0.063)%以及LDH活性抑制了(35.1±0.267)%。另外,通过流式细胞仪(FACS)测定结果得出咖啡果皮粗提物对细胞具有保护作用。

【Abstract】 The Yunnan Arabica coffee has been tropical characteristic industry in Yunnan province. The coffee husks were removed and discarded during the dehulling process, however, coffee husks could be used as potential sources of anthocyanins. This paper was aim to investigate anti-oxidative injuries effect of crude extracts from Yunnan Arabica coffee husk for HUVEC cells. The results indicated that the extracts possessed potentially protective effects for cells that underwent oxidative injuries due to 400 μmol/L of hydrogen peroxide(H2O2) treatment, compared with the H2O2 treatment alone cells group, 400 mg/L of the extract increased the activity of superoxide dismutase(SOD) in cells by(9.6±0.35)%, and decreased the concentration of malondialdehyde(MDA) and the activity of lactate acid dehydrogenase(LDH) by(23.0±0.063)% and(35.1±0.267)%, respectively. In addition, the fluorescence activated cell sorter(FACS) results by Annexin V-FITC/PI showed that the extract possess potential anti-apoptosis activity,which apoptosis induced by hydrogen peroxide(H2O2). These results suggested that the extract had anti-oxidative properties.
