酶辅助提取沉香叶多酚及其抗氧化性研究

【摘要】 以沉香叶为原料,采用酶辅助有机溶剂浸提法提取沉香叶多酚,探讨了酶种类、乙醇浓度、酶解温度、酶解pH值、酶添加量、底物质量浓度及酶解时间对多酚得率的影响,在单因素基础上,运用正交试验优化提取工艺;采用清除自由基法评价多酚提取物的抗氧化性。结果表明,纤维素酶辅助乙醇法提取沉香叶多酚的效果最好,最佳提取条件为:乙醇浓度60%(v/v),酶解温度50℃,酶解pH5,酶添加量200 U/g、底物质量浓度5 g/100 mL,时间2 h,此条件下多酚的得率为4.96%(w/w);沉香叶多酚提取物具有较强的抗氧化性,清除ABTS自由基、DPPH自由基和·OH自由基的半数有效浓度(EC50)值分别为(0.025±0.002)、(0.106±0.006)、(0.12±0.001)mg/mL。

【Abstract】 Orthogonal design was applied to optimize the of polyphenols fromLignum Aquilariae Resinatum leaves by ethanol and enzymatic assistant extraction. The yield of polyphenols was investigated with discussing the varities of enzymes, ethanol concentration, enzymolysis temperature, pH, addition level of enzyme, substrate mass concentration and time. Best results for the extraction of polyphenols were achieved by ethanol and Cellulase assistant extraction. The optimal extraction conditions were determined as follows: 60% ethanol as the extraction solvent, 50 ℃, pH5, 200 U/g enzyme adsition, 5 g/100 mL substrate mass concentration and 2 h extraction time. Under these conditions, the extraction yield of polyphenols fromLignum Aquilariae Resinatum leaves was 4.96%. And the extracted polyphenols presented strong antioxidant activity. The median effective concentrations(EC50) values of ABTS radical,DPPH radical and hydroxyl radical scavenging rate was(0.025±0.002) mg/mL,(0.106±0.006) mg/mL,(0.12±0.001) mg/mL, respectively.
