响应面法优化林蛙干皮胶原蛋白提取工艺

【摘要】 为提高林蛙皮胶原蛋白的提取率,并对其特征进行初步鉴定。在优化林蛙皮脱色、脱脂预处理工艺的基础上,采用响应面法优化胃蛋白酶酶解制备林蛙干皮胶原蛋白的工艺,并采用紫外扫描、红外扫描和凝胶电泳技术对所获林蛙干皮胶原蛋白进行初步特征鉴定。结果发现林蛙干皮最佳预处理工艺条件为:室温下用含有1.0%H2O2的0.15 mol/L NaOH脱色12 h,10%正丁醇按料液比1:75脱脂5 h;酶解最佳工艺条件为:胃蛋白酶添加量为5%、酶解时间为3.5 h、酶解温度为50℃、酶解料液比为1:45;在此参数条件下,实际林蛙胶原蛋白的提取率则为94.89%,并且林蛙皮胶原蛋白具有Ⅰ型胶原蛋白的特征。该研究为林蛙皮胶原蛋白的提取提供了实验依据,为林蛙皮综合利用奠定基础。

【Abstract】 In order to improve the extraction rate of collagen from forest frog dry skin and identify the characteristics,it was optimized the pre-treatment process of forest frog dry skin bleaching and degreasing.Then,response surface methodology was used to optimize the pepsin hydrolysis process of collagen extraction.The UV scanning,infrared scanning and gel electrophoresis techniques were used to preliminarily characterize the forest frog skin collagen.As a results,the best pre-treatment conditions were:decolorization of 12 h at room temperature and with 0.15 mol/L NaOH(contain 1.0%H2O2),degreasing with 10%n-butanol at the ratio of material to liquid of 1:75 for 5 h.The optimum enzymatic hydrolysis conditions were that 5%pepsin,hydrolysis time of 3.5 h,enzymolysis temperature of 50 °C,ratio of solid to liquid of 1:45.Under this condition,the actual collagen extraction rate was 94.89%.Moreover,forest frog skin collagen has the characteristics of type Ⅰ collagen.This study provided the basis for the collagen extraction from forest frog and laid foundation for the utilization of the forest frog.
