双酶法水解发酵豆渣制备大豆多肽的研究

【摘要】 以豆渣为原料,首先采用黑曲霉对豆渣进行发酵,使豆渣中的蛋白初步水解,再采用中性蛋白酶和木瓜蛋白酶对豆渣进行双酶水解,制备大豆多肽。单因素和正交试验结果表明:发酵提高了豆渣蛋白的水解度,并且在发酵和双酶同时处理条件下,最佳工艺为中性蛋白酶与木瓜蛋白酶比例3:1、酶浓度6000 U/g、底物浓度6%、p H7.0、最适温度50℃、水解时间6 h,在该条件下,水解度为42.83%。

【Abstract】 With bean dregs as raw materials,using Aspergillus niger fermented soybean residue,preliminary hydrolyzed protein in soybean residue,then the flavor protease and papain were applied to hydrolyze soybean residue to prepare soybean peptides.The single factor and orthogonal experiment results showed that the fermentation improved the hydrolysis degree of soybean protein and with the condition of fermentation and double enzyme,The optimal condition were as follows:flavor protease and papain ratio 3:1,enzyme concentration 6000 U/g,substrate concentration 6%,p H7.0,optimum temperature 50 ℃,hydrolysis time of 6 h,the maximum DH was 42.83%.
