顺铂联合原花青素对宫颈癌Hela细胞生长抑制作用
Study on the effects of cisplatin combined with grape seed proanthocyanidins on the cell proliferation of human cervical cancer Hela cells
【摘要】 目的探讨顺铂联合葡萄籽原花青素对宫颈癌Hela细胞的生长抑制作用。方法 2015年10月—2016年1月体外培养宫颈癌Hela细胞,分为对照组、顺铂组、原花青素组及顺铂联合原花青素组,48、72 h后,倒置显微镜下观察细胞形态学变化,CCK8法检测细胞增殖抑制率。三组及以上计量资料比较采用单因素方差分析,两两比较采用LSD法;计数资料比较采用χ2检验,P<0.05为差异有统计学意义。结果顺铂及原花青素对Hela细胞的增殖抑制呈现剂量和时间相关性,随着药物浓度的升高和作用时间的延长,Hela细胞的增殖抑制率升高。联合用药组9、18、37μmol/ml的顺铂联合340μmol/ml的原花青素分别作用48 h后Hela细胞的增殖抑制率分别为75.7%、76.1%、76.2%,72 h后Hela细胞的增殖抑制率分别为77.0%、78.0%、78.9%。两者联合用药组细胞增殖抑制率高于单独用药组,差异均有统计学意义(均P<0.05)。结论不同浓度顺铂、原花青素对Hela细胞增殖均有抑制作用,原花青素对Hela抑制作用显著,顺铂联合原花青素对宫颈癌细胞Hela有协同抑制作用。
【Abstract】 Objective To study the inhibition effect of cisplatin combined with grape seed proanthocyanidins on the proliferation of Hela cells.Methods Hela cells were cultured in vitro and divided into control group,cisplatin group,grape seed procyanidins group and combined group from October 2015 to January 2016.The morphological changes of cells were observed by inverted microscope and cell inhibition rate were detected by CCK8 method.Measurement data was processed by variance analysis,pairwise comparison was used by LSD.Count data was used by chi square test,P<0.05 was considered statistically significant.Results The inhibition effect of cisplatin on Hela cells was in a time and dose dependent manner.With the increase of drug concentration and the extension of effect time,the proliferation inhibition rate of Hela cell increased.The concentrations of cisplatin were 9,18,37μmol/ml in combination group,they respectively reacted with grap seed pranthocyanidins whose concentration was 340 μmol/ml,after 48 h,the proliferation inhibition rates of Hela cell were 75.7%、76.1%、76.2%,respectively.After 72 h,the proliferation inhibition rates of Hela cell were 77.0%,78.0% and 78.9% respectively.The cell proliferation inhibition rate in the combination group was significantly higher than that in cisplatin group and grape seed proanthocyanidins group(all P<0.05).Conclusion Various concentrations of cisplatin and grape seed proanthocyanidins have inhibitory effect on Hela proliferation,cisplatin combined with grape seed proanthocyanidins have synergistic inhibition effect on cervical cancer Hela cells.
