蜜楝不同部位绿原酸积累变化分析研究

【摘要】 利用高效液相色谱法分析测定了不同生长时期蜜楝叶、花和果实中绿原酸的含量。色谱条件为:色谱柱为Inertsil ODS-3C18(4.6×150 mm,5μm)柱,以乙腈-0.2%磷酸溶液(8∶92)为流动相,流速1.0 m L/min,检测波长327 nm,柱温30℃。绿原酸在30.4μg/m L~152μg/m L范围内呈良好的线性关系,Y=27.335X-7.67(R2=0.999 8),平均加样回收率100.77%,RSD为1.58%(n=6)。分析结果表明,在蜜楝植物不同部位中绿原酸的含量不同,其中叶含量在4.69 mg/g~8.90 mg/g之间,花中含量在14.59 mg/g~27.30 mg/g之间,果实中含量在8.79 mg/g~16.70 mg/g之间,在同一部位绿原酸的积累随生长时间的不同而表现出一定的变化。研究结果表明,蜜楝的花及果实可作为提取制备绿原酸成分的一种资源,具有一定开发利用价值。

【Abstract】 The contents of chlorogenic acid in Evodia lenticellata Huang in different times were determined by using HPLC. The chromatographic separation was carried out using Inertsil ODS-3 C18column(4.6 mm×150 mm,5 μm),the mobile phase was acetonitrile :0.2 % phosphoric acid(8 ∶ 92),the detection wavelength was set at327 nm,the temperature was 30 ℃,and the flow rate was 1.0 m L/min. The calibration curve was linear in the range of 30.4 μg/m L-152 μg/m L and the regression equation for chlorogenic acid was Y=27.335X-7.67(R2=0.999 8),the average recovery was 100.77 % with RSD of 1.58 %(n=6). The result showed that there are some difference in the content of chlorogenic acid among different periods,the content of chlorogenic acid were range between 4.69 mg/g to 8.90 mg /g in the leaf,14.59 mg/g to 27.30 mg /g in the flower and 8.79 mg/g to 16.70 mg /g in the fruit. The accumulation of chlorogenic acid in the same parts of different time shows certain regularity. The study showed that the flower and fruit of E. lenticellata Huang can be used as a kind of resource to extract chlorogenic acid.
