浒苔蛋白酶法改性提高原料抗氧化性能工艺优化
Process Optimization of Protein Enzymatic Modification to Improve Enteromorpha Oxidation Resistance
【摘要】 以浒苔粉为原料制成浒苔蛋白溶液,采用定向酶解技术改性浒苔蛋白,通过碱性蛋白酶处理的方式,基于单因素,浒苔蛋白酶解产物的还原力为指标,采用二次通用旋转组合试验设计方案,优化碱性蛋白酶酶解改性浒苔蛋白关键工艺条件。结果表明,控制浒苔粉溶液的料液比为1:30,经优化后的碱性蛋白酶酶解改性浒苔蛋白关键工艺参数组合为:碱性蛋白酶使用量为600 U/g浒苔粉,调节酶解液pH为8.0,在50℃下水浴提取时间为90 min,以此工艺条件组合结合其他单因素条件进行验证性扩大试验,改性后的酶解液中的浒苔蛋白酶解液的还原力最高,为0.792。
【Abstract】 With enteromorpha powder as raw material,the enteromorpha protein solution was modified by alkaline protease successively.Based on single factor,the key processing parameters of alkaline protease were optimized by quadratic universal rotary assembly experiment.The reducing power of enzymatic hydrolysate was used as the metric.The results showed that when the condition of solid-liquid ratio was 1:30,the optimized process parameters of alkaline protease are as follows:usage amount of alkaline protease 600 U/g,pH 8.0,temperature 50 ℃ and water incubator time 90 min.In this condition,expanding test were conducted combining with other single factors,the reducing power of hydrolysate was the highest,which can be up to 0.792.
