HPLC法测定杜仲茶中车叶草苷含量的研究
Study on HPLC Method for Asperuloside Detecting in Eucommiae Tea
【摘要】 采用C18柱,流动相为甲醇+水+磷酸(66+870+1),流速1.5m L/min,检测波长237nm,柱温40℃左右,进样量20μL。通过研究检测线性关系、稳定性、精密度、重复性、加标回收率等,建立了HPLC法测定杜仲茶中车叶草苷含量的检测方法。结果表明:杜仲茶车叶草苷在0.0126-0.1134mg·mL-1之间线性良好(R2=0.9993),平均回收率为102.9%,测得车叶草苷含量为1.48%。该方法准确性高,重现性好,是一种快速、简便检测杜仲茶中车叶草苷含量的方法。
【Abstract】 A C18 column(Mightysil RP-18 GP Aqua 150-4.65μm) was adopted as stationary phase, mobile phase of methanol+water+phosphate(66+870+1), the flow rate of 1.5m L/min, detection wavelength of 237 nm, column temperature 40℃, 20μL of incoming sample amount. By studying the detection of linear relation, stability, precision,reproducibility, standard addition recovery rate, etc., established a HPLC method for determination of asperuloside from eucommia tea. Results show that eucommia tea asperuloside content had a good linear relation(R~2=0.9993) in the range of 0.0126-0.1134mg/mL, the average recovery was 102.9%, asperuloside content is 1.48%. The method is of high accuracy, good reproducibility and it is a fast and simple method of testing asperuloside content of Eucommia tea.
