乳腺癌患者血清与肿瘤组织中VEGF、ES表达和微血管密度的关系

【摘要】 目的探讨乳腺癌患者术前血清中血管内皮生长因子(VEGF)、血管内皮抑素(ES)表达与术后组织VEGF、ES、微血管密度(MVD)表达的相关性。方法采集221例乳腺癌患者术前血清,用ELISA酶联免疫吸附法测定VEGF和ES含量,用免疫组化SP法检测患者术后乳腺癌组织中VEGF、ES、MVD表达。分析乳腺癌患者术前血清VEGF、ES表达与组织VEGF、ES、MVD表达的相关性。结果乳腺癌患者血清VEGF与组织VEGF显著正相关(P<0.01,RS=0.746);血清VEGF与组织MVD显著正相关(P<0.05,R=0.141);血清VEGF与组织ES显著正相关(P<0.01,RS=0.375);血清ES与组织ES显著正相关(P<0.01,RS=0.386);组织VEGF与组织ES显著正相关(P<0.01,RS=0.178)。结论乳腺癌患者外周循环血中VEGF和ES来自肿瘤细胞分泌;VEGF增多导致肿瘤新生血管增多,微血管密度增加;在肿瘤细胞大量分泌VEGF的过程中,可能对抗血管生成的ES也相应增加,只是VEGF过量表达抵消了ES的血管抑制作用,使总的平衡趋向于血管生成。

【Abstract】 Objective To explore the correlation between VEGF and ES expressions in serum and VEGF,ES and MVD expressions in the tissue before surgery in the patients of breast cancer. Methods The serum of 221 breast cancer patients was collected. ELISA method was used to determine the levels of VEGF and ES. IHC- SP method was used to determine the expressions of VEGF,ES and MVD expressions in cancer tissue. The correlation was analyzed between VEGF and ES expressions in serum and VEGF,ES and MVD expressions in tissue before surgery. Results Serum VEGF is significantly and positively relevant with tissue VEGF in the patients( P < 0. 01,RS= 0. 746),the serum VEGF was significantly and positively relevant with tissue MVD( P < 0. 05,R = 0. 141),serum VEGF was significantly and positively relevant with tissue ES( P< 0. 01,RS= 0. 375),serum ES was significantly and positively relevant with tissue ES( P < 0. 01,RS=0. 386) and tissue VEGF was significantly and positively relevant with tissue ES( P < 0. 01,RS= 0. 178).Conclusion VEGF and ES in peripheral blood are from tumor cellular secretion in the patients of breast cancer. VEGF increase induces probably the progression of tumor angiogenesis. VEGF over- expression neutralizes the vascular inhibition of ES to enable the general balance tending to angiogenesis.
