高效液相色谱-蒸发光散射检测法(HPLC-ELSD)测定小儿清热宁颗粒中胆酸的含量

【摘要】 目的:建立小儿清热宁颗粒中胆酸的含量测定方法;方法:样品制备采用甲醇超声30 min提取,色谱条件为色谱柱Venusil XBP C18(L)柱(4.6 mm×250 mm,5μm),流动相甲醇-1%冰醋酸溶液(75∶25,v/v),流速1.0 m L/min,柱温为25℃;检测器漂移管温度控制为40℃,载气气体流速为2.0 L/min,增益为4。结果:胆酸的工作曲线为lg A=1.582 lg C+2.972 9(r=0.999 4),进样量在0.502~2.51μg范围内呈良好的线性关系,回收率的RSD为1.969 6%。阴性对照无干扰。结论:所建立的含量测定方法专属性强,可用于小儿清热宁颗粒的质量控制。

【Abstract】 Objective:To establish an HPLC-ELSD method for determination of cholic acid in Xiaoer Qingrening Granule. Methods:Samples were prepared by ultrasonic extraction of methanol for 30 min. Venusil XBP C18(L)(4. 6 mm × 250 mm,5 μm)column was used with methanol- 0. 1% phosphoric acid solution(75:25,v / v) with the mobile phase at 1. 0 m L / min. The temperature of column was set at 25 ℃ and drift tube was 40 ℃. The flow rate of gas was maintained at 2. 0 L / min. Results:The linearity was obtainedfrom 0. 502 ~ 2. 51 μg( r = 0. 999 4). The average recovery was 100. 532 1% with RSD = 1. 969 6%. Conclusion:The method is specific,and can be used for quality control of Xiaoer Qingrening Granule.
