亚硒酸钠对胡萝卜愈伤组织诱导形成的影响
Effect of Sodium Selenite on the Induction of Carrot Callus
【摘要】 选取新鲜胡萝卜的形成层为试验材料,研究亚硒酸钠对胡萝卜愈伤组织诱导形成的影响.在添加2.0mg/L 2,4-D和0.8mg/L 6-BA的MS培养基中分别加入0,0.5,1.0,2.0mg/L的亚硒酸钠诱导培养愈伤组织,观察愈伤组织的诱导情况并计算愈伤组织的诱导率,同时测定抗氧化能力的相关指标,包括总抗氧化能力、GSH-PX活力、SOD活力、H2O2浓度、MDA含量.实验结果表明:亚硒酸钠浓度为0.5mg/L时,胡萝卜愈伤组织被有效诱导,愈伤组织生长状况最佳,抗氧化能力显著提高.
【Abstract】 Fresh Carrot cambium selected as experimental materials to study the effect of sodium selenite on the induction of Carrot callus.Added the concentration of sodium selenite 0,0.5,1.0,2.0mg/L,respectively,into MS medium with 2.0mg/L 2,4-D and 0.8mg/L 6-BA to observe the induction of callus,to calculate the induction rate of callus and to determinate the antioxidant capacity,namely,total antioxidant capacity,GSH-PX and SOD activity,H2O2 and MDA concentration.The results show that the optimal concentration of sodium selenite added in MS medium is 0.5mg/L.Carrot callus are induced effectively,growth condition of callus are improved,the antioxidant capacity of callus are significantly increased.
