掌叶半夏提取物对宫颈癌荷瘤小鼠脾脏免疫的作用
Effects of Pinellia extracts on immune responds of spleen in cervical cancer-bearing mice
【摘要】 目的观察掌叶半夏提取物(Pinellia extracts,PE)对HPV+TC-1荷瘤小鼠T细胞免疫应答的作用。方法宫颈癌荷瘤小鼠建模后随分成4组:机对照组(生理盐水,0.2 mL/d,灌胃)、PE低剂量组(10 mg/kg,0.2mL/d,灌胃)、PE高剂量组(20 mg/kg,0.2 mL/d,灌胃)、CDDP组(4 mg/kg,0.2 mL/3 d,腹腔注射)。给药期间测瘤、描绘肿瘤生长曲线,21天后处死小鼠并计算抑瘤率,流式细胞术检测脾脏CD4+和CD8+T细细胞以及Th1、Th2、Treg和Th17亚群的比例。结果抑瘤率:CDDP组(29.82%)>PE高剂量组(21.73%)>PE低剂量组(8.23%);PE高剂量组和CDDP组CD4+和CD8+T细胞比例较阴性对照组均显著增多(P<0.01);PE两组Th1比例显著升高、Th2和Th17比例显著下降(呈现剂量依赖性),Treg亚群分泌TGF-!减少,而FOXP3水平增高。结论 PE能通过诱导CD4+T细胞向Th1型分化,抑制Th1/Th2漂移,改善Treg和Th17的比例,发挥抗肿瘤免疫作用。
【Abstract】 Objective To observe the role of Pinellia extracts(PE)in T cells immune response of the C57BL/6 mice bearing HPV+TC-1 tumor. Methods After randomization and administration in cervical cancer-bearing mice,tumor volume was calculated and the growth of tumor was completed.After transplanting subcutaneous tumor,the mice were randomly divided into 4 groups:control group(normal saline,0.2 mL/d,gavage),PE-low group(10 mg/kg,0.2 mL/d,gavage),PE-high group(20 mg/kg,0.2mL/d,gavage),and CDDP group(4 mg/kg,0.2 mL/3d,peritoneal injection).Besides,the jobs of calculating the volume of the tumors and drawing tumor growth curve were done.The tumor inhibitory rate was calculated after continuous dosing for 21 days,and the percentages of CD4+,CD8+ T cells and Th1,Th2,Treg and Th17 subgroup in the spleen was detected by flow cytometry. Results The order of tumor inhibitory rate was:CDDP group(29.82%)>PE high dose group(21.73%)>PE low dose group(8.23%).Compared with control group,the percentage of CD4+ and CD8+ T cells of PE high dose group and CDDP group were significantly increased(P< 0.01).In both PE groups,the proportions of Th1 and Th17 were significantly increased,while Th2 significantly decreased(in dose-dependent manner).Treg subsets secreted less TGF-β,while FoxP3 level increased. Conclusions Through inducing the differentiation of CD4 + T cells to Th1 model,inhibiting the drift of Th1/Th2 and regulating the proportion of Treg and Th17,PE could play an important role in anti-tumor immunity.
