日粮添加硒对热应激条件下蛋鸡相关生理生化指标及HSP70 mRNA表达的影响
Effects of adding Se to diet on some physiological and biochemical indexes and HSP70 mRNA expression of laying hens under heat stress
【摘要】 在热应激条件下,对产蛋高峰期的海兰蛋鸡分别饲喂添加有0 mg/kg、0.3 mg/kg、0.5 mg/kg、0.7 mg/kg硒的基础日粮,分别在试验第0天(试验开始前一天)、第1天、第7天、第14天和第21天的时候,对各组鸡的血清进行采样,测定样品中的三碘甲状腺原氨酸(T3)、皮质醇(CORT)含量;对各组鸡的胸肌、肝脏进行采样,测定各组织热应激蛋白70(HSP70)基因的表达量。结果显示:热应激对蛋鸡血清T3含量无显著影响,但显著提高了热应激第1天和第7天血清CORT的含量;日粮中添加硒可在一定程度上提高热应激条件下蛋鸡血清T3含量,降低CORT含量;热应激极显著地提高了海兰蛋鸡胸肌、肝脏组织中HSP70 mRNA的表达量;日粮中添加硒能极显著地降低蛋鸡胸肌、肝脏HSP70 mRNA的表达量。
【Abstract】 Under heat stress Hailan hens at egg-laying peak stage were fed on basal diets added respectively with 0 mg/kg,0.3 mg/kg,0.5 mg/kg and 0.7 mg/kg Se,their sera were sampled respectively on the day before test and the 1st,7th,14 th and 21 st days of test,and determined in 3,5,3 ’-triiodothyronine(T3)and Cortisol(CORT)contents;Also the hens’tissues were determined in heat shock protein 70(HSP70) expression level.The results showed that heat stress had not a significant effect on the serum T3 content but significantly increased the serum CORT content on the 1st and 7th days of test;To a certain extent adding Se to basal diet could increase the serum T3 content and decrease the serum CORT content under heat stress;Heat stress very significantly increased the HSP70 mRNA expression levels in the hens’pectoral muscles and livers,but adding Se to basal diet could decrease the HSP70 mRNA expression levels very significantly.
