不同食管病变组织中TGFβ1、α-SMA、smad4、VEGF蛋白表达及MVD比较
Comparison of expression of TGFβ1,α-SMA,smad4,VEGF and comparison of MVD in different esophageal lesions
【摘要】 目的比较正常食管组织、低级别食管上皮内瘤变、高级别食管上皮内瘤变、早期食管癌和进展期食管癌组织中TGFβ1、α-SMA、smad4、VEGF蛋白表达及微血管密度(MVD)。方法选取正常食管组织标本20份(NC组),低级别食管上皮内瘤变组织标本30份(LGIEN组),高级别食管上皮内瘤变组织标本50份(HGIEN组),早期食管癌组织标本25份(CIS组)和进展期食管癌组织标本25份(SCC组),采用免疫组化法检测各组标本中的TGFβ1、α-SMA、smad4、VEGF蛋白,用CD34标记血管内皮细胞测算各组MVD。结果从NC组到SCC组,食管间质成纤维细胞α-SMA、TGFβ1、VEGF阳性表达率和MVD逐渐升高(P均<0.05);上皮细胞smad4蛋白阳性表达率逐渐下降(P均<0.05),上皮细胞TGFβ1阳性表达率先升后降,CIS组最高(P均<0.05)。结论随着正常食管组织向进展期食管癌组织转变,其中间质成纤维细胞的表型和功能发生改变,TGFβ1/smad4蛋白通路和血管新生可能在其中发挥重要作用。
【Abstract】 Objective To detect and compare the changes of transform growth factor β1( TGFβ1),α-SMA,smad4 protein,vascular endothelial growth factor( VEGF) and microvessel density( MVD) in the normal esophageal tissues,lowgrade intraepithelial neoplasia( LGIEN),high-grade intraepithelial neoplasia( HGIEN),early esophageal cancer and advanced esophageal cancer tissues. Methods Twenty cases of normal esophageal tissue specimens( NC group),30 cases of LGIEN specimens( LGIEN group),50 cases of HGIEN specimens,25 cases of early esophageal cancer specimens( CIS group) and 25 cases of advanced esophageal cancer specimens( SCC group) were selected. The expression of TGFβ1,α-SMA,smad4 and VEGF protein in each group was detected by immunohistochemistry,and the MVD was determined by Antibody CD34. Results From NC group to SCC group,the positive expression rates of α-SMA,TGFβ1 and VEGF and MVD were gradually increased( all P < 0. 05); the positive expression rate of smad4 protein was gradually decreased( all P < 0. 05); and the positive expression rate of TGFβ1 was first increased and then decreased,and the CIS group was the highest( all P < 0. 05). Conclusions In the course of the development of esophageal cancer from normal esophagus to advanced stage,the phenotype and function of esophageal stromal fibroblasts change,and TGF β1 / smad4 protein pathway and angiogenesis may play an important role in the protein pathway.
