高效液相色谱法测定不同品种番木瓜果实中维生素C含量

【摘要】 通过优化实验条件,使用高效液相色谱法对海南省种植范围较广的几种番木瓜(中白、大青、小青)维生素C(Vc)含量进行测定。选取3个番木瓜品种成熟果实,通过HPLC方法测定Vc的标准溶液线性、精密度、回收率等,并比较该法与国标方法的重复性。结果显示,获得线性回归方程y=35.41x-4.16(r=0.999),Vc在60~300μg/m L的浓度范围线性关系良好,方法回收率为98.56%~103.21%,精密度RSD为0.69%;测得中白、大青、小青果肉Vc含量分别为49.12、43.38、46.31 mg/hg。HPLC法结果重现性RSD为0.68%;2,6-二氯靛酚滴定法结果重现性RSD为5.45%,表明HPLC法是检测番木瓜等深颜色水果Vc含量可靠的方法。

【Abstract】 In order to develop an optimal method for isolating and analyzing Vc in breeds of papaya which were widely planted. extracting solution, chromatographic conditions were improved and optimized on the basis of former reported High Performance Liquid Chromatography(HPLC) methods. The results showed that The calibration curves of Vc was in good linearity over the ranges of 60~300 μg/m L(r=0.999),and the average recovery rates was 100.75%(RSD=0.69%). The content of Vc in "Zhongbai", "Daqing", "Xiaoqing" is 49.12, 43.38, 46.31 mg/100 g. The method of HPLC is better in repeatability(RSD=0.68%)than 2,6-dichlorindophenol titration(RSD=5.45%). Therefore HPLC could be used for the determination of Vc in papaya and other fruits which had high level of Vc.
