【摘要】 通过阿式肠杆菌和萎缩芽孢杆菌样品制备的经验方法,在JSM-6700F电镜下得到形态完整高清晰的图像.培养获得高纯度的菌,采用双固定、磷酸缓冲液清洗、丙酮脱水、多次表面镀铂等对样品进行处理.结果是电镜观察样品形态的丰富度高;样品扫描图像有丰富的立体感;分辨率高图像更清晰.为其他生物扫描电镜观察提供了参考.更多还原

【Abstract】 To get high-resolution and shape integral graphics of Enterobacter Asburiae and Bacillus Atrophaeus using JSM-6700 F.The method is that,after acquisition of pure microorganism by precisely control the culture condition in media,we prepared the sample with double fixation,phosphate buffer solution rinse,acetone dehydration,and multiple surface platinum plating.High-resolution and shape integral graphics were achieved using SEM after the preparation.The micororganism were in richmorphological diversity and stereoscopic impression.Our results would be benefit to other researchers using SEM.更多还原
