急性冠脉综合征患者血小板反应蛋白-1基因多态性分析

【摘要】 目的研究中国人群血小板反应蛋白-1(TSP-1)基因A2210G(N700S)多态性与急性冠脉综合征(ACS)的相关性。方法采用病例-对照研究,入选2005年6月~2010年11月在江苏大学附属武进医院和江苏省人民医院住院的412例ACS患者及同期337例非冠状动脉粥样硬化性心脏病(冠心病)者作为对照组,应用聚合酶链反应-限制性片段长度多态性分析的方法检测TSP-1 A2210G多态性,采用logistics回归分析TSP-1基因多态性分布与ACS的相关性。结果 TSP-1基因A2210G多态性AG基因型在ACS组和对照组的分布无显著性差异(1.5%vs.0.6%,P=0.43),未检测到GG纯合子。G等位基因频率在ACS组和对照组分别为0.7%、0.3%(P=0.43)。多元Logistic回归分析显示,TSP-1基因AG基因型与ACS无显著相关性(OR=2.36;95%CI:0.46~12.48;P=0.46)。结论 TSP-1基因700N→S变异在中国人群中发生频率低,且与ACS的发病无显著相关性。

【Abstract】 Objective To analyze the correlation between polymorphism of the thrombospondin-1(TSP-1) gene A2210G(N700S) and acute coronary syndrome(ACS) in the Chinese population. Methods A case-control study was conducted. By polymerase chain reaction and restriction fragment length polymorphism analysis, the TSP-1 A2210 G polymorphism was analyzed in 412 patients with ACS(ACS group) and 337 control subjects free from coronary artery disease(control group) from Affiliated Wujin Hospital of Jiangsu University between June 2005 and November 2010. Results The frequencies of AG genotype were 1.5% and 0.6% in ACS group and control group, respectively(P=0.43). None of the homozygotes were detected for the G allele. The prevalence of the G allele was not significantly different between ACS group and control group(0.7% vs. 0.3%, P=0.43). Result of multivariate logistic regression analysis showed that AG genotype of TSP-1 gene had no significant correlation with ACS(OR=2.36;95%CI:0.46~12.48;P=0.46). Conclusion The TSP-1 N700 S variant shows a low prevalence in the Chinese population and fails to associate with an altered risk of ACS.
