一种新型可注射口腔根管充填糊剂的遗传毒性实验评价

【摘要】 目的:对自主研发的新型可注射根填充糊剂进行体外细胞遗传毒性研究。方法:首先采用MTT法确定材料对中国仓鼠肺成纤维细胞(CHL)的半数细胞生长抑制浓度(IC50),根据IC50,选用最高浓度60%,中、低浓度按照两倍等比稀释原则分别为30%和15%,利用三种浓度作用CHL细胞24h和48h,评价可注射根填充糊剂对细胞染色体的损伤情况。结果:阳性对照物中非代谢活化组的丝裂霉素C诱发产生的畸变率为13.5%(24h)和13.5%(作用48h),代谢活化组的环磷酰胺诱发产生的畸变率为11.0%,与阴性对照组比较具有统计学意义上的显著性差异(P<0.01)。自主研发的可注射根填充糊剂低、中、高浓度分别作用CHL细胞24h和48h,细胞畸变率均小于5%,与阴性对照组比较,未见统计学意义上的显著性差异,即自制可注射根填充糊剂对CHL细胞染色体无遗传毒性。

【Abstract】 Objective:To Investigate the in vitro genetic toxicity of a novel injectable root filling paste.Methods:Firstly,the MTT assay was applied to determine the half cell growth inhibitory concentration(IC50) on the chinese hamster lung into fibroblast(CHL).Secongly,according to the IC50,we chose the highest concentration of 60%,and middle and low concentration in accordance with the principle of twice the geometric dilution respectively 30%and 15%.By contacting with the CHL cells for 24 h and 48 h,the cell chromosome damage was evaluated for the injectable root filling paste.Results:In non-metabolic activation,the induced distortion rate of positive control of mitomycin C was 13.5%(24h) and 13.5%(48h),and In metabolic activation group,the distortion rate induce by cyclophosphamide was 11.0%,which were all statistically significant difference(P < 0.01) with the negative control group.When interacted with the CHL cells for 24 h and 48 h,the cell distortion rate conducted by the self-fabricating injectable root canal filling paste with low,medium and high concentrations were less than 5%.The was no significant difference(P<0.01) as compared with the negative control group,indicating that the self-fabricating injectable root canal filling paste had no genetic toxicity on CHL cell chromosome.
