彩叶植物红叶石楠离体再生体系的研究
In Vitro Regeneration Research in Photinia serrulata cv‘hongye’ with Colored-leaf
【摘要】 选取红叶石楠幼茎和顶芽为外植体,以MS为基本培养基,添加不同浓度的6-BA和NAA激素进行离体培养,比较各种激素组合对红叶石楠初代培养、不定芽分化、生根培养的作用和影响。结果表明:初代培养的最佳培养基为MS+1.5 mg/L 6-BA+0.05 mg/L NAA;诱导不定芽分化的最适培养基为MS+2.0 mg/L 6-BA+0.2 mg/L NAA,诱导率达到86.7%;生根的最佳培养基为1/2 MS+3.0 mg/L NAA,生根率达到92.7%;组培苗移栽成活率为94.6%。
【Abstract】 An efficient method has been developed for In Vitro regeneration of Photinia serrulata cv‘hongye’in order to protect and utilize its genetic resources. The apical buds and stems were used as the explants to investigate the effect of different plant regulators on inducement of explants, differentiation of adventitious buds and rooting by using MS medium with different 6-benzylamino purine(6-BA) and1-naphthaleneacetic acid(NAA). The result of this study showed that the optimum inducement of explants medium was MS added with 1.5 mg/L 6-BA and 0.05 mg/L NAA, the optimal differentiation of adventitious buds and rooting medium were MS with 2.0 mg/L 6-BA and 0.2 mg/L NAA and 1/2MS+3.0 mg/L NAA respectively. The differentiation rate and rooting percentage were 86.7% and 92.7% respectively. The survival rate of transplants was reached 94.6%. The establishment of tissue culture system on Photinia serrulata cv ‘hongye’will preserve its genetic resources and provide technical support for its industrial production.
