川芎嗪对糖尿病大鼠降血糖及肾脏保护作用的研究

【摘要】 目的 探讨川芎嗪对糖尿病大鼠降血糖及肾脏保护作用。方法 将经链脲佐菌素诱导成功的24只糖尿病大鼠随机分为川芎嗪组,阳性对照组,模型组,分别以川芎嗪200mg/(kg·d)、缬沙坦25mg/(kg·d)、生理盐水10ml/(kg·d)灌胃,另取8只正常大鼠为正常对照组,生理盐水10ml/(kg·d)灌胃,连续给药6周。给药后每周称量大鼠体重,第2、4、6周检测大鼠空腹血糖;第6周代谢笼法测定24h尿量,并检测尿蛋白总量(U—TP)与尿微量白蛋白(U—ALB);第6周末心脏取血,处死大鼠,酶联免疫法检测血清中胰岛素的含量;取双肾,计算肾肥大指数,光镜下观察肾脏病理切片。结果 模型组大鼠的血糖、24h尿量、U—TP、U—ALB、肾重、肾肥大指数较正常组均有显著升高(P<0.01或P<0.05),血清胰岛素显著降低(P<0.05),身体机能明显下降,肾脏病理明显加重;经川芎嗪治疗6周后,大鼠血糖、24h尿量、U—TP、肾重与肾肥大指数明显下降(P<0.05),但血清胰岛素无明显升高(P>0.05),身体机能显著改善,肾脏病理状态有所减轻。结论川芎嗪对糖尿病大鼠有一定的降血糖与肾脏保护作用,但其降血糖机制并非通过增加胰岛素的分泌

【Abstract】 Objective To research the effect of blood sugar reduction and renal protection of ligustrazine on diabetic rats.Methods 24 diabetic rats successfully induced by streptozotocin(STZ) were randomly divided into ligustrazine group,positive control group,model group(DM group),and treated with ligustrazine 200 mgkg-1·d-1,valsartan 25 mg·kg-1·d-1,physiological saline 10 mg·kg-1·d-1 respectively.8 healthy rats were served as normal control group(NC group,physiological saline 10ml/kg·d).All rats were treated for 6 weeks continuously.The rats weighed weekly after treating,the blood sugar was measured on 2th,4th,6th week.The 24 h total urine was measured by metabolic cage,total urine protein(U-TP) and total micro albumin(U-ALB) were measured on 6th week;The blood sample was obtained from the heart by puncture,and rats were executed at the end of 6th week,,serum insulin(INS) was detected by Enzyme linked immunosorbent assay(ELISA).Kidney hypertrophy index was calculated and pathological changes of kidneys were also observed under light microscope.Results In DM group,blood glucose,total urine,U-TP and U-ALB,kidney weight and kidney hypertrophy index were significantly higher than that in NC group(P<0.01,P<0.05),the serum INS was significantly lower,the physical conditions were significantly decreased,and renal pathological status was significantly serious.After ligustrazine intervention,blood glucose,total urine and U-TP,kidney weight and kidney hypertrophy index were significantly lower(P<0.05),but serum INS had no significant change,the physical conditions and renal pathological status were significantly improved.Conclusion Ligustrazine has some effect on reducing blood sugar and protecting kidney on diabetic rats,and its mechanism of reducing blood sugar was irrelevant with INS.
