【摘要】 目的探讨Rab25蛋白(Ras-associated blinging protein)和磷酸肌醇3-激酶(Phosphatidylinositol 3-Kinase,PI3K)在肾透明细胞癌(clear cell renal cell carcinoma,ccRCC)中的表达及临床意义。方法选取43例ccRCC癌组织标本及23例癌旁正常肾组织标本,分别应用免疫组织化学染色及反转录聚合酶链反应(RT-PCR)法检测Rab25、PI3K表达及mRNA相对表达情况,分析二者表达的相关性及其与临床病理特征的关系。结果 Rab25及PI3K阳性表达主要定位于胞膜或胞浆,癌旁正常肾组织多不着色。ccRCC癌组织中Rab25、PI3K阳性表达率及mRNA相对表达量均明显高于癌旁正常肾组织(P<0.05);Rab25、PI3K在ccRCC中的表达呈正相关(r=0.487,P<0.01),且二者的表达与肿瘤临床分期、淋巴结转移、病理分级密切相关(P<0.05)。结论 Rab25和PI3K可能与ccRCC的发展、侵袭、转移有紧密联系,且二者存在协同作用。更多还原

【Abstract】 Objective To investigate the expression of Rab25 protein and PI3 K protein in clear cell renal cell carcinoma( ccRCC) occurrence,development and invasion,and to analyze their correlation. Methods 43 cases of ccRCC specimens underwent surgical treatment,and 23 specimens of para-carcinoma tissues were obtained. Immunohistochemistry was used to detect the expression of Rab25 protein and PI3 K protein in the two groups; RT-PCR was used to detect the expression of Rab25 and PI3 K mRNA in the two groups; and then the difference and correlation of the expression between Rab25 with PI3 K,and the correlation of the expression with the tumor clinicopathological parameters were analyzed. Results The results of immunohistochemistry showed that both Rab25 protein and PI3 K protein had been stained on cytomembrane or cytoplasm,but not in para-carcinoma tissues,and both of them had a significantly higher positive expression rate in ccRCC tissues than that in para-carcinoma tissues( P < 0. 05). RT-PCR results showed that the relative quantity of mRNA from Rab25 and PI3 K also had significant differences between ccRCC tissues with para-carcinoma tissues( P < 0. 05). The expression of Rab25 protein had a positive correlation with the expression of PI3 K protein( r = 0. 487,P < 0. 01),and they were significantly associated with the clinical stage,lymphnode metastasis and pathological grade( P < 0. 05). Conclusion An osculating relationship may exist between the high expression of Rab25 protein and PI3 K protein with the development,invasion and metastasis of ccRCC and therefore,the expression of Rab25 protein may have a positive correlation with that of PI3 K protein in those aforementioned processes.更多还原
