慢性酒精暴露对不同性别Wistar大鼠前额叶皮质htr3a mRNA表达水平及觅药行为的影响
The effect of chronic ethanol exposure on htr3a mRNA expression in prefrontal cortex and ethanol-seeking behavior in different sex Wistar rats
【摘要】 目的 探讨慢性酒精暴露对不同性别Wistar大鼠前额叶皮质htr3a mRNA表达水平及觅药行为的影响。方法 将24只雄性和24只雌性成年健康Wistar大鼠随机分为4组,雄性酒精组、雄性对照组、雌性酒精组、雌性对照组,每组12只,分别予以酒精或生理盐水处理15 d。采用条件性位置偏爱模型的条件性位置偏爱分值评价大鼠的觅药行为。采用实时荧光定量PCR检测大鼠前额叶皮质htr3a mRNA表达水平。结果 雄性酒精组及雌性酒精组条件性位置偏爱分值、前额叶皮质htr3a mRNA表达水平均显著高于雄性对照组及雌性对照组(P<0.01),不同性别大鼠酒精组和对照组条件性位置偏爱分值、前额叶皮质htr3a mRNA表达水平比较差异均无显著性(P>0.05)。雄性和雌性酒精组条件性位置偏爱分值与前额叶皮质htr3a mRNA表达水平呈显著正相关(r=0.441,P<0.05)。结论 Wistar大鼠慢性酒精暴露后前额叶皮质htr3a mRNA表达水平升高,且与酒精觅药行为呈正相关;雄性与雌性Wistar大鼠慢性酒精暴露诱导的觅药行为、前额叶皮质htr3a mRNA的表达水平无显著差异。
【Abstract】 Objective To explore the effect of chronic ethanol exposure on htr3 a mRNA expression in prefrontal cortex(PFC) and ethanol-seeking behavior in different sex Wistar rats.Methods Twenty-four male and 24 female Wistar rats were randomly assigned to four groups of 12 ones each;male ethanol,male control,female ethanol and female control group,each group was exposed to ethanol or saline for 15 days.Their ethanol-seeking behaviors were assessed using conditioned place preference(CPP) scores.The htr3 a mRNA expression levels in PFC were detected using real-time fluorescence quantitative PCR.Results CPP scores and htrSa mRNA expression levels in PFC were higher in both male and female ethanol group than in male and female control group(P<0.01),there were no significant differences in those between different sex ethanol and control group(P>0.05).CPP scores in both male and female ethanol group were positively related to the htr3 a mRNA expression levels in PFC notably(r=0.441,P<0.05).Conclusion After chronic ethanol exposure the htr3 a mRNA expression levels in the PFC in Wistar rats increase,which are positively correlated with CPP scores;there are no significant differences in ethanol-seeking behavior induced by chronic ethanol exposure and the htr3 a mRNA expression levels between male and female Wistar rats.
