BDNF在急性应激所致小鼠空间学习记忆功能改变中的作用
Role of BDNF in the Spatial Learning-memory Function of Mice Induced by Acute Stress
【摘要】 目的研究急性应激对小鼠空间学习记忆功能及脑内BDNF表达的影响。方法采用足底电击急性应激动物模型,通过Morris水迷宫实验检测试小鼠空间学习记忆能力;以免疫组织化学方法检测脑源性神经营养因子(BDNF)在海马和前脑皮层的表达。结果与正常对照组小鼠比较,急性应激组小鼠在水迷宫实验中的逃避潜伏期明显缩短(P<0.05,P<0.01),而在目标象限的停留时间显著延长(P<0.05);急性应激组小鼠海马CA1、CA3区,齿状回和前脑皮层BDNF的表达明显增加(P<0.05,P<0.01)。结论急性应激导致小鼠空间学习记忆功能增强,这可能与海马和前脑皮层BDNF表达的上调有关。
【Abstract】 Objective To study the effects of acute stress on the ability of spatial learning and memory and the expression of brain derived neurotrophic factor(BDNF) in the hippocampus(HP) and prefrontal cortex(PFC) of mice. Methods Using animal model mice induced by single-factor acute stressor(foot shock). The ability of spatial learning and memory of mice was measured using Morris water maze, and the expressional changes of BDNF in mice brain were detected by immunohistochemical method. Results Compared with control group mice, the escape latency of the acute stress group mice were shorter(P<0.05, P<0.01), and the swimming time of the platform quadrant of the acute stress group mice were significantly increased in Morris water maze(P<0.05). The expression of BDNF in the CA1,CA3 region and dentate gyrus of HP and PFC were significantly increased in stress group mice(P<0.05, P<0.01), compared with control group mice. Conclusion The results indicated that acute stress induced a significant increase in spatial learning and memory ability, and it may be nearly related to the up-regulation of BDNF expression in HP and PFC.
