从桦褐孔菌中首次分离ganodecochlearin A

【摘要】 〔目的〕对研究桦褐孔菌的羊毛脂烷型三萜成分进行研究。〔方法〕采用回流提取法对桦褐孔菌子实体进行提取,待所得提取物均匀混悬于水后,依次采用石油醚、乙酸乙酸、正丁醇进行萃取;将乙酸乙酯萃取物分别采用反复的硅胶柱色谱、ODS开放柱色谱及制备型HPLC进行化学成分的分离,对分离得到的化合物采用1H-NMR、13C-NMR、DEPT、HSQC、HMBC、HHCOSY、NOESY及HRMS等波谱数据进行结构鉴定。〔结果〕从桦褐孔菌乙酸乙酯萃取物中分离得到羊毛脂烷型化合物ganodecochlearin A(1),它的结构被鉴定为22R,25-环羊毛甾-7,9-二烯-3β,24S-二醇。〔结论〕首次从桦褐孔菌中分离出化合物ganodecochlearin A,并在综合波谱解析的基础上对其1H-NMR和13C-NMR数据进行了正确的归属。

【Abstract】 〔Objective〕To search for the bioactive constiruents of Inonotus obliquus. 〔Methods〕The powered materials was extracted with 95% Et OH under reflux and the extracts was fractioned by petroleum ether, ethyl acetate and n-butanol successively to 4 parts. The ethyl acetate extracts was futher isolated by silica gel, ODS and HPLC chromatograph to obtain the triterpenoid. Its structure was identified by physicochemical properties,1H-NMR,13C-NMR, DEPT, HSQC, HMBC, HHCOSY,NOESY and HRMS. 〔Results〕A lanostane-tape triterpene( ganodecochlearin A( 1)) was isolated from the ethyl acetate extracts of I. obliquus. Its structure was established as 22 R, 25-epoxylanost-7, 9-diene-3β, 24S-diol. 〔Conclusion 〕Ganodecochlearin A is isolated from I. obliquus for the first time. The position of its1H-NMR and13C-NMR data are elucidated correctly on the basis of detailed analysis of the 1D and 2D NMR spectroscopy and HRMS.
