硫化氢对百草枯中毒大鼠急性肺损伤及炎症介质的影响

【摘要】 目的观察硫化氢(H2S)对百草枯(PQ)中毒大鼠急性肺损伤(ALI)及炎症介质的影响。方法将30只雄性SD大鼠随机分为3组,每组10只,正常对照组(A组),PQ中毒组(B组),NaHS干预组(C组)。B、C组采用PQ灌胃(80mg/kg)复制大鼠PQ中毒肺损伤模型,B组同时腹腔注射生理盐水(6ml/kg),每日两次,C组PQ染毒后,同时腹腔注射NaHS(1.4umol/kg),每日两次;A组采用等量生理盐水灌胃,同时腹腔注射生理盐水(6ml/kg),每日两次。72h后取血检测肿瘤坏死因子-а(TNF-а)、白细胞介素-6(IL-6)、白细胞介素-8(IL-8)的含量;在光学显微镜下观察肺组织病理学改变,进行肺损伤病理评分。结果 B组中TNF-а、IL-6、及IL-8含量明显高于A组(P<0.01);C组中TNF-а、IL-6、及IL-8含量较B组明显减低(P<0.05);B组大鼠肺损伤病理评分高于A组(P<0.01);C组大鼠肺损伤病理评分较B组减低(P<0.01)。结论 H2S具有抑制百草枯中毒大鼠炎症介质表达,减轻大鼠急性肺损伤作用。

【Abstract】 Objective To investigate the effect of hydrogen sulfide(H2S) on acute lung injury(ALI) and inflammatory factors in rats with paraquat poisoning. Methods 30 male SD rats were randomly divided into 3 groups,10 rats in each group,normal control group(A group),PQ poisoning group(B group),NaHS intervention group(C group). B,C,group of rats were made into the model of lung injury by pouring PQ(80mg/Kg) into the stomach,B group was injected with normal saline(6ml/kg) at the same time,two times a day. C group rats were exposed to PQ,while intraperitoneal injection of NaHS(1.4μmol/kg),two times a day. In group A,the same amount of normal saline was used to fill the stomach,normal saline(6ml/kg) was injected by intraperitoneal at the same time,two times a day. Determinating the concentration of tumor necrosis factor-а(TNF-а),interleukin-6(IL-6),interleukin-8(IL-8) in peripheral blood after 72 hours. Observing the pathological changes of lung tissue,counting lung injury score under optical microscope. Results In group B,TNF-а,IL-6,and IL-8 were significantly higher than that of A group(P<0.01). In group C,TNF-а,IL-6,and IL-8 was significantly lower than that of group B(P<0.05). The lung injury score in B group was significantly higher than that in A group(P <0.01). The lung injury score in the C group was significantly lower than that in the B group(P<0.01). Conclusion H2S can inhibit the expression of inflammatory mediators in rats with paraquat poisoning,and reduce the acute lung injury in rats.
