桑色素对全氟辛酸诱导小鼠肝损伤的保护作用
Protective Effect of Morin on Perfluorooctanoic Acid-Induced Liver Injury in Mice
【摘要】 目的探讨桑色素(morin,Mor)对全氟辛酸(perfluorooctanoic acid,PFOA)诱导小鼠肝损伤的保护作用。方法将24只雄性小鼠按随机数字表法分为4组:对照组(等容量生理盐水)、PFOA组(PFOA 10 mg·kg-1·d-1)、Mor组(Mor 75mg·kg-1·d-1)和PFOA+Mor组(PFOA 10mg·kg-1·d-1+Mor 75mg·kg-1·d-1),每组6只。连续灌胃14d。分别检测血清谷丙转氨酶、谷草转氨酶和碱性磷酸酶的活性,肝组织匀浆中丙二醛、C-反应蛋白、白细胞介素-6的水平,超氧化物歧化酶、过氧化氢酶和环氧化酶-2的活性。结果与对照组比较,PFOA组、PFOA+Mor组小鼠血清谷丙转氨酶、谷草转氨酶、碱性磷酸酶活性及PFOA组小鼠肝组织匀浆中丙二醛、C-反应蛋白、白细胞介素-6水平和环氧化酶-2活性均明显升高,PFOA组、PFOA+Mor组小鼠肝组织匀浆中超氧化物歧化酶、过氧化氢酶活性均明显降低(均P<0.05);与PFOA组比较,Mor组、PFOA+Mor组小鼠血清谷丙转氨酶、谷草转氨酶和碱性磷酸酶活性,肝组织匀浆中丙二醛、C-反应蛋白、白细胞介素-6水平和环氧化酶-2活性均明显降低,肝组织匀浆中超氧化物歧化酶、过氧化氢酶活性均明显升高(均P<0.05)。结论桑色素对PFOA诱导的小鼠肝损伤具有保护作用,其机制可能与抑制氧化应激和炎症反应有关。
【Abstract】 Objective To investigate the protective effect of morin(Mor)on perfluorooctanoic acid(PFOA)-induced liver injury in mice.Methods Twenty-four male mice were randomly divided into four groups:control group(same volume of normal saline),PFOA group(PFOA 10mg·kg-1·d-1),Mor group(Mor 75mg·kg-1·d-1)and PFOA+Mor group(PFOA 10mg·kg-1·d-1+Mor 75 mg·kg-1·d-1),with 6 mice in each group.After treatment for 14 consecutive days,serum activities of alanine transaminase(ALT),aspartate transaminase(AST)and alkaline phosphatase(ALP),hepatic contents of malondialdehyde(MDA),and hepatic activities of superoxide dismutase(SOD),catalase(CAT),C-reactive protein(CRP),interleukin-6(IL-6)and cyclooxygenase-2(COX-2)were measured in all groups.Results Compared with control group,liver injury indicators ALT,AST and ALP,inflammatory response markers CRP,IL-6and COX-2,and lipid peroxidation product MDA increased,but antioxidant enzymes SOD and CAT decreased in PFOA group(P<0.05).Compared with PFOA group,simultaneous supplementation of Mor decreased serum activities of ALT,AST and ALP,reduced hepatic contents of MDA,CRP,IL-6and COX-2,and increased hepatic activities of SOD and CAT(P<0.05).Conclusion Mor protects against PFOA-induced liver injury in mice through anti-oxidation and anti-inflammation.
