NAD~+抑制脂多糖诱导海马炎症因子的表达
NAD~+ suppressed the expressions of inflammation factors in the hippocampus induced by lipopolysaccharide
【摘要】 采用1 mg·kg-1脂多糖(LPS)腹腔注射30 min后进行烟酰胺腺嘌呤二核苷酸(NAD+,腹腔注射100、300mg·kg-1)干预,并在LPS注射3和6h后,采用实时定量PCR方法检测海马中白细胞介素-1β(IL-1β)和肿瘤坏死因子-α(TNF-α)的mRNA表达量,探讨NAD+干预对LPS诱导的C57BL/6雄性小鼠海马炎症因子IL-1β和TNF-αmRNA表达量变化的影响。结果表明:LPS诱导海马中IL-1β和TNF-αmRNA表达量明显升高;LPS注射3h后,NAD+(100、300mg·kg-1)均显著抑制了LPS诱导的海马中IL-1βmRNA表达量的升高;LPS注射6h后,NAD+(100mg·kg-1)明显抑制了LPS诱导的海马中TNF-αmRNA表达量的升高。说明NAD+抑制LPS诱导的海马炎症因子的表达,提示NAD+可用于由脑部炎症引起的脑疾病早期干预治疗。
【Abstract】 Lipopolysaccharide(LPS,1mg·kg-1)was injected intraperitoneally,and mice were treated withβ-Nicotinamide adenine dinucleotide(NAD+,100,300mg·kg-1)30min after the LPS injection.The changes in expressions of interlukin-1β(IL-1β)and tumor necrosis factor-α(TNF-α)mRNA in hippocampus at 3 and 6 hafter LPS injection,respectively,were measured by quantitative real-time PCR.The effect of NAD+on the expressions of LPS-induced inflammation factors,IL-1 β and TNF-α,in the hippocampus of C57BL/6 mice was investigated.Results showed that:LPS led to the dramatic elevation of IL-1β and TNF-α mRNA in hippocampus.NAD+(100,300 mg·kg-1)markedly inhibited the augment of IL-1 β mRNA at 3 hafter LPS injection.NAD+(100 mg·kg-1)significantly suppressed the elevation of TNF-αmRNA at 6 hafter LPS administration.The data demonstrated that NAD+suppressed the expressions of inflammation factors in hippocampus induced by LPS,implying the application prospect of NAD+in early-stage treatment of encephalitis or other brain diseases caused by cerebral inflammation factors.
