miR-26a靶向作用于COX-2及其对宫颈癌细胞增殖和侵袭的影响
The effect of miR-26a on proliferation and invasion of human cervical cancer cells by targeting COX-2
【摘要】 目的探讨miR-26a靶向作用于环氧合酶-2(COX-2)及其对宫颈癌Siha细胞株增殖和侵袭的影响。方法 Real-time PCR法检测21例患者宫颈癌组织及对应的癌旁组织中mi R-26a的表达,用Western bolt法检测COX-2的表达。利用脂质体将pre-mi R-26a或anti-mi R-26a瞬时转染至宫颈癌Siha细胞,real-time PCR分析mi R-26a与COX-2基因表达的调控关系。Western blot法检测转染后细胞中COX-2的表达水平,同时采用CCK-8和克隆形成实验检测转染后Siha细胞的增殖和侵袭情况。结果 mi R-26a在宫颈癌组织中的表达明显低于癌旁组织(P<0.01),而COX-2在宫颈癌组织中的表达明显高于癌旁组织(P<0.01)。转染pre-mi R-26a后,宫颈癌Siha细胞株内COX-2的表达量显著降低,而转染anti-mi R-26a后则显著上升(P<0.01)。CCK-8和克隆形成实验结果显示,过表达mi R-26a能明显抑制宫颈癌Siha细胞的增殖和侵袭。结论 mi R-26a抑制宫颈癌Siha细胞的增殖和侵袭可能与下调COX-2的表达相关。
【Abstract】 Objective To study the effect of miR-26 a on proliferation and invasion of cervical cancer Siha cells by targeting cyclooxygenase-2(COX-2). Methods The expression of miR-26 a in 21 cases of cervical cancer tissues and corresponding para-carcinoma tissues was detected by real-time PCR, and Western blot method was employed to detect the expression of COX-2 protein. The pre-miR-26 a or anti-miR-26 a were transiently transfected into cervical cancer cell Siha with Lipofectamine. CCK-8 and clone formation experiments were employed to determine the proliferation and invasion ability of transfected Siha cells. Results The expression level of mi R-26 a was significantly lower in cervical cancer tissues than in para-carcinoma tissues(P <0.01),but the expression level of COX-2 was significantly higher in cervical cancer tissues than in para-carcinoma tissues(P <0.01). The expression levels of COX-2 gene and protein were significantly decreased in Siha cells after pre-miR-26 a was transfected, but significantly increased in Siha cells after anti-mi R-26 a was transfected(P <0.01). CCK-8 and clone formation experiment revealed that the proliferation and invasion abilities of Siha cells were markedly inhibited by the overexpression of mi R-26 a. Conclusion The inhibition of MiR-26 a on proliferation and invasion abilities of Siha cells might be related to down-regulating the expression of COX-2.
