对氯苯丙氨酸抑制野百合碱诱导的大鼠肺动脉和右心室重构

【摘要】 目的研究对氯苯丙氨酸(4-Chloro-DL-phenylalanine,PCPA)对野百合碱(monocrotaline,MCT)诱导的大鼠肺动脉和右心室重构的影响。方法建立MCT诱导的大鼠肺动脉高压(pulmonary arterial hypertension,PAH)模型,腹腔注射给予不同剂量PCPA。检测肺动脉压力、右心指数等血流动力学指标。HE染色观察肺动脉及心肌形态学的改变。Western Blot方法检测右心室基质金属蛋白酶13(matrix metalloproteinases 13,MMP-13)的表达。结果 MCT诱导的大鼠肺动脉压力、右心指数、肺动脉血管壁厚度、心肌细胞的损伤及右心室中MMP-13蛋白表达水平比对照组明显增加,而PCPA则剂量依赖性的降低以上各个指标。结论 PCPA抑制MCT诱导的大鼠肺动脉和右心室的重构可能与降低MMP-13蛋白表达相关。

【Abstract】 Objective To study the effect of 4-Chloro-DL-phenylalanine(PCPA) on monocrotaline(MCT)-induced remodeling of pulmonary artery and right ventricle in rats. Methods Pulmonary arterial hypertension models were induced by monocrtaline and PCPA(intraperitoneal injection of PCPA). The pulmonary arterial pressure and right ventricle hypertrophy index were measured, the change of pulmonary artery and myocardial morphology was observed by HE straining, the expression of matrix metalloproteinases-13(MMP-13) in right ventricle was detected by Western blot. Results The pulmonary artery pressure, right ventricle hypertrophy index, pulmonary vascular wall thickness, the damage of myocardial cells and expression level of MMP-13 protein in right ventricle were increased significantly compared to the normal control, but significantly decreased by PCPA with dose-dependent manner. Conclusion The inhibition of the pulmonary arterial remodeling and right ventricular hypertrophy by PCPA might be related to the decrease of MMP-13 protein.
