miR-101及Runt相关转录因子1在肺癌细胞系A549中的作用
Role of miR-101 and Runt-related transcription factor 1 in lung cancer cell line A549
【摘要】 目的研究miR-101及其下游靶基因Runt相关转录因子1(RUNX1)在肺癌细胞系A549中的作用。方法 Western Blot及Real Time PCR检测miR-101和RUNX1在A549中的表达情况,上调或下调miR-101后A549中RUNX1的表达变化,绘制生长曲线及细胞克隆实验检测A549细胞的增殖变化,transwell验证A549细胞迁移的变化。结果在A549细胞,miR-101表达水平下调,RUNX1表达水平上调;miR-101抑制A549细胞的增殖及迁移;RUNX1促进A549细胞的增殖及迁移。结论 miR-101通过抑制RUNX1的转录表达抑制肺癌细胞系A549的增殖及迁移。
【Abstract】 Objective To study the role of miR-101 and Runt-related transcription factor 1(RUNX1) in the lung cancer cell line A549. Methods Western blot and Real Time PCR were used to detect the expression of miR-101 and RUNX1 to explore the regulatory role of miR-101 on RUNX1 expression, the proliferation of A549 cells was detected by drawing growth curve and cell clone,the migration of A549 cells was detected by transwell. Results In A549 cells, the expression level of miR-101 was down-regulated, and the expression level of RUNX1 was up-regulated. Mi R-101 inhibited the proliferation and migration of A549 cells, and RUNX1 promoted the proliferation and migration of A549 cells. Conclusion miR-101 inhibits the proliferation and migration of lung cancer cell line A549 by inhibiting the transcription and expression of RUNX1.
