【摘要】 目的探讨miR-551b-3p在胃癌中的表达变化情况,及其对胃癌细胞功能的影响。方法用real-time PCR的方法检测60例胃癌组织及其对应的癌旁组织中miR-551b-3p的表达量,使用miR-551b-3p模拟物(miR-551b-3p mimic)转染胃癌细胞系HGC-27,CCK-8法检测细胞增殖;划痕愈合实验检测细胞迁移;Transwell法检测细胞侵袭。结果 1)miR-551b-3p在胃癌癌症组织中的表达明显低于癌旁组织(P<0.05)。2)过表达miR-551b-3p mimic可以明显减弱胃癌细胞系HGC-27的增殖、迁移和侵袭能力。结论 miR-551b-3p在胃癌组织中低表达,并且抑制胃癌细胞系HGC-27的增殖、迁移和侵袭,可能与胃癌发生发展密切相关。更多还原

【Abstract】 Objective To investigate the expression and change of miR-551b-3p in gastric cancer and its effect on the function of gastric cancer cells. Methods miR-551b-3p was detected by RT-PCR in 60 cases of gastric cancer samples and control tissues. HGC-27 cell line was infected by miR-551b-3p mimic,then CCK-8 assay was used to determine the influence of miR-551b-3p on HGC-27 cell proliferation; Wound-healing assay was used to determine cell migration; Transwell assay was used to determine cell invasion. Results 1) The expression of miR-551b-3p in gastric tumor tissues was significantly lower than that of the adjacent tissues( P < 0. 05). 2) HGC-27 cell proliferation,migration and invasion significantly decreased when the cell was infected by mimic-miR-551b-3p. Conclusions miR-551b-3p expresses less in gastric cancer tissues and inhibits gastric cancer line HGC-27 proliferation,migration and invasion. miR-551b-3p is closely associated with the development of gastric cancer.更多还原
