牛卵泡闭锁与颗粒细胞凋亡基因表达研究

【摘要】 颗粒细胞凋亡与卵泡闭锁是颗粒细胞功能基因及凋亡相关基因的表达结果,通过手术法获取三种不同直径的牛卵泡(小型卵泡<2 mm;中型卵泡2~6 mm;大型卵泡>6mm),分离卵泡颗粒细胞并提取总RNA,以研究不同直径卵泡中颗粒细胞相关基因的表达。结果表明,牛卵巢上小型卵泡颗粒细胞中Fshr的表达显著低于中型和大型卵泡(P<0.05),而中型和大型两者之间差异不显著(P>0.05);中型卵泡和小型卵泡颗粒细胞中CYP19的相对表达显著低于大型卵泡(P<0.05);中型卵泡和小型卵泡颗粒细胞中促细胞凋亡基因BAX的表达显著低于大型卵泡(P<0.05),而在中型和大型卵泡颗粒细胞中抑制细胞凋亡基因BCL2的表达显著低于小型卵泡(P<0.05),且随着卵泡的增大而下调;Casapase 8与Caspase3的表达模式相同,其表达量均随着卵泡直径的增加显著上升(P<0.05)。随着卵泡生长与直径增加,卵泡内部分颗粒细胞虽然在继续增殖,部分颗粒细胞已经开始凋亡,卵泡逐渐进入闭锁阶段。

【Abstract】 Apoptosis of granulosa cells and follicular atresia are the expressions of functional genes and apoptosis related genes in granulosa cells which were isolated by dissection from three types of bovine ovarian follicle,namely,small follicle with the diameter less than 2 mm,medium follicle with the diameter ranged from 2~6mm and large follicle with the diameter more than 6mm.The granulose cells RNA was extracted.Gene’s relative expression was measured with Real-Time quantitative PCR after the reverse transcription.The results showed the expression of Fshr in the small follicles in bovine ovary was significantly lower than that of medium and large follicles(P<0.05),and there was no significant difference between the previous two groups(P>0.05).The expression of CYP19 in medium and small follicles was significantly lower than that of larger follicle(P<0.05).The expression of pro-apoptotic gene Bax in granulosa cells of small and medium-sized was significantly lower than that of large size follicle(P<0.05),while the expression of anti-apoptotic gene BCL2 with lower abundance in large and medium-sized follicle decreased with the growth of follicle(P< 0.05).Casapase 8and Caspase 3had the same expression pattern,which expression increased with the growth of follicle(P<0.05).It was concluded that with the growth and the diametric increase of bovine follicle,follicle entered the atresia phase with apoptosis of some granulosa cells,although some genes functioned normally in partial granulosa cells in follicle.
