白术对脑老化小鼠认知能力及相关信号蛋白表达的影响

【摘要】 目的探讨白术对D-半乳糖致脑老化小鼠学习记忆能力的影响。方法建立D-半乳糖脑老化动物模型,用被动回避实验检测小鼠学习记忆能力,用Western blot检测海马区突触素、蛋白激酶C、环一磷酸腺苷反应元件结合蛋白的表达水平。结果被动回避实验:脑老化组的潜伏期[(149.75.±83.92)s],对照组[(260.17±42.81)s]两组比较,P<0.01有显著性差异;错误次数[(5.75±1.51)次],对照组[(1.08±0.43)次]两组比较P<0.01,有显著性差异;而RAM低、中、高剂量组的潜伏期[(231.92.±55.71)s,(257.75±43.11)s,(269.33±33.79)s]与脑老化组比较(P<0.05,P<0.01)有显著性差异,中、高剂量组的错误次数[(1.58±0.53)次,(0.83±0.50)次]与脑老化组比较P<0.01有显著性差异。Western blot实验:与对照组相比,脑老化组海马Syn、PKC、CREB的表达显著降低,而RAM组可显著提高Syn、PKC、CREB的表达。结论白术可显著改善脑老化小鼠的学习记忆能力,其机制可能与上调海马Syn、PKC、CREB的表达有关。

【Abstract】 Objective To investigate the effects of rhizoma atractylodis macrocephalae(RAM) on learning-memory in D-galactose-induced brain aging mice. Methods The brain aging model was induced by S.C D-galactose. Learning-memory ability was tested by passive avoidance test and Morris water maze test,and the expression of synapsin(Syn), protein kinase C(PKC) and c AMP-response element binding protein(CREB) in hippocampus were examined by Western blot. Result Passive avoidance test: the latency in brain aging group(149.75.17 ± 83.92)s) significantly decreased, and the number of errors(5.75±1.51) significantly increased compared with the control group( latency:(260.17±42.81) s; number of errors(1.08 ± 0.43),(P<0.01)).The latency low dose, middle dose and high dose RAM group((231.92.±55.71)s,(257.75±43.11)s,(269.33±33.79)s) significantly increased, and the number of errors in middle dose and high dose RAM group((1.58±0.53),(0.83±0.50)) significantly decreased compared with brain aging group(P<0.05,P<0.01). Western blot test: the expression of Syn, PKC and CREB in hippocampus of brain aging group was significantly weakened than that of the control group. Conclusion RAM could significantly improve the ability of learning and memory in brain aging mice. Its effects might be related to the increase of the expression of Syn, PKC and CREB in hippocampus.
