二甲双胍联合异羟肟酸对骨肉瘤细胞增殖凋亡的影响
Effect of Combining Metformin and Suberoylanilide Hydroxamic Acid on the Proliferation and Apoptosis of Osteosarcoma Cells
【摘要】 目的研究二甲双胍(Met)联合异羟肟酸(SAHA)应用对人骨肉瘤细胞MG-63增殖和凋亡的影响。方法 MTS法分别检测二甲双胍、SAHA和二甲双胍联合SAHA对MG-63细胞生长的抑制作用;平板克隆实验检测二甲双胍联合SAHA对MG-63细胞克隆形成能力的影响;流式细胞仪检测二甲双胍联合SAHA对MG-63细胞凋亡的影响;Western Blot检测联合用药后细胞周期和凋亡相关蛋白的变化。结果二甲双胍和SAHA分别对MG-63细胞生长有抑制作用,二甲双胍联合SAHA应用对MG-63细胞生长的抑制作用更加显著;二甲双胍联合SAHA应用与单药相比能够明显降低MG-63细胞克隆形成率,并且促进细胞凋亡;联合用药后P27,P21,Cyclin D1,Mcl-1,XIAP,Bim,Cytochrome C蛋白发生明显改变。结论二甲双胍联合SAHA应用与单药相比能够显著抑制人骨肉瘤MG-63细胞的增殖,促进其凋亡,可见两药联用对肿瘤细胞的杀伤具有协同性。
【Abstract】 Objective To investigate the effect of combining metformin and suberoylanilide hydroxamic acid( SAHA)on the proliferation and apoptosis of human osteosarcoma MG-63 cells. Methods MG-63 cells were treated with metformin and SAHA for 72 h. The proliferation of MG-63 was tested by MTS assay. The capability of colony formation of MG-63 was assessed by clonogenicity assay. The cell apoptosis was detected by flow cytometry. The cell cycle and apoptosis associated proteins were assessed by Western Blot. Results Metformin or SAHA could inhibit the proliferation of MG-63 cells,respectively,and the cell proliferation was inhibited more obviously after combination of theses two drugs. Moreover,combination of two drugs could inhibit colony formation and promote cell apoptosis more significantly. P27,P21,Cyclin D1,Mcl-1,XIAP,Bim,Cytochrome C were significantly changed after the combination therapy. Conclusion Combination of metformin and SAHA could inhibit proliferation and colony formation and promote apoptosis more effectively in MG-63 cells.
