miR-483过表达对小鼠记忆功能的影响
Impact of miR-483 overexpression on mice memory retention
【摘要】 目的探讨miR-483过表达是否影响小鼠记忆功能。方法选取C57BL/6低龄(3月龄)及高龄(13月龄)的野生型及miR-483转基因鼠,进行新物体识别实验检测记忆能力。PCR筛选实验鉴定转基因阳性小鼠后,分析两种小鼠记忆能力的差异。运用实时定量PCR方法检测各组小鼠脑中miR-483表达水平,以及记忆相关分子在脑中的表达趋势。通过Western blot方法检测小鼠脑中记忆相关蛋白的表达水平。结果低龄转基因型小鼠与低龄野生型小鼠新物体识别次数差异具有统计学意义,低龄野生型鼠记忆能力更强;低龄转基因小鼠脑中细胞外调节蛋白激酶(extracellular regulated protein kinases,ERK)表达量高于低龄野生型小鼠。结论在低龄小鼠中,miR-483过表达导致记忆损伤,同时引起脑部ERK表达水平的上调。
【Abstract】 Objective To analyze the impact of miR-483 overexpression on mice memory function. Methods Young( 3 month) and aged( 13 month) mice of wild-tpye or miR-483 transgenic group were selected. Mice in each group were tested by novel object recognition and the results were recorded. PCR screening was used to determine the mouse with wild-type or transgenic and then analyzed the differences in the behavior experiment. Real-time PCR was used to detect brain miR-483 and some key molecules on memory retention expression levels. Western blot was performed to detect the expression level of memory-related protein in mouse brain. Results Young WT mice got greater capacity of memory. The ERK expression in brain of young TG mice was higher than young wild-type ones. Conclusion In young mice,miR-483 overexpression leads to a worse ability on the memory retention and the upregulation of ERK expression in brain.
