Snail1基因启动子区组蛋白修饰在前列腺癌细胞EMT发生中的作用
Snail1 gene promoter region histone modification in the role of EMT on prostate cancer cells
【摘要】 目的探讨Snail1基因启动子区的组蛋白修饰与前列腺癌DU145细胞发生上皮间质转化(epithelial-mesenchymal transition,EMT)现象的关系。方法 RT-PCR、Western blot及IF法验证TGF-β1诱导前列腺癌细胞发生EMT过程中相关标记物的变化;利用Ch IP试验来筛查Snail1启动子区组蛋白修饰及RNA聚合酶Ⅱ的富集区域。结果诱导前列腺癌发生EMT过程中,仅有DU145细胞EMT相关标记物的变化明显(P<0.05);DU145细胞中Snail1经TGF-β1诱导后出现明显上调的现象;Snail1基因启动子区域的P4位点存在H3K4ME3及RNA聚合酶Ⅱ的富集。结论 TGF-β1通过促使Snail1基因启动子区P4位点的H3K4ME3富集导致Snail1表达上调,促进了前列腺癌DU145细胞EMT现象的发生。
【Abstract】 Objective To explore the relationship between histone modification of Snail1 promoter and EMT in prostate cancer DU145 cells.Methods RT-PCR,Western blot and IF were used to detect the changes of relevant markers in EMT induced by TGF-β1,and Ch IP test was used to screening histone modification and RNA polymerase Ⅱ enrichment area of snail1 promoter.Results Markers of EMT changed significantly in DU145 cells treated with TGF-β1(P<0.05);Expression of Snail1 was obviously increased;P4 site of Snail1 promoter existed enrichment areas of H3K4ME3 and RNA polymerase Ⅱ.Conclusion TGF-β1could up-regulate Snail1 expression through enrichment of H3K4ME3 and RNA polymerase Ⅱ in P4 site of Snail1 promoter,promoting EMT in DU145 cells.
