高效液相色谱法同时测定阿归养血颗粒中芍药苷和阿魏酸的含量
High-performance liquid chromatography for simultaneous determination of paeoniflorin and ferulic acid in Agui Yangxue granules
【摘要】 目的:建立高效液相色谱法测定阿归养血颗粒中芍药苷和阿魏酸的含量。方法:采用Hypersil BDS C18柱(250mm×4.6 mm,5μm);流动相为乙腈-0.1%磷酸溶液(15∶85);检测波长:阿魏酸为323nm,芍药苷为230nm;流速:1.0m L·min-1;柱温:室温。结果:此方法下芍药苷和阿魏酸分别在9.245~147.9μg/m L、6.432~102.9μg/m L与峰面积呈良好的线性关系;平均回收率和RSD分别为98.9%和1.7%、97.5%和1.6%。结论:本研究所建立的HPLC法准确、快速、灵敏度高,可用于阿归养血颗粒中芍药苷和阿魏酸的含量测定。
【Abstract】 Objective: To investigate the high- performance liquid chromatography( HPLC) method for the simultaneous determination of paeoniflorin and ferulic acid in Agui Yangxue granules. Methods: HPLC was performed on a Hypersil BDS C18column( 250 mm × 4. 6mm,5μm) with a mobile phase of acetonitrile- 0. 1% phosphoric acid solution( 15∶ 85) at a wavelength of 323 nm for ferulic acid and 230 nm for paeoniflorin,a flow rate of 1. 0 ml / min,and a column temperature of room temperature. Results: Paeoniflorin and ferulic acid showed a good linear relationship between concentration and peak area within the ranges of 9. 245- 147. 9 μg / m L and 6. 432 ~ 102. 9μg / m L,respectively,and the average recovery rates were 98. 9%( RSD = 1. 7%) and 97. 5%( RSD = 1. 6%),respectively. Conclusion: The HPLC method established in this experiment is accurate,time- saving,and highly sensitive and can be used for the determination of paeoniflorin and ferulic acid in Agui Yangxue granules.
