内皮型一氧化氮合酶活性的调节与心血管疾病的研究进展
Research progress in the regulation of endothelial nitric oxide synthase activity and cardiovascular diseases
【摘要】 内皮型一氧化氮合酶(endothelial nitric oxide synthase,e NOS)是合成一氧化氮(nitiric oxide,NO)起主要作用的酶,e NOS和NO在调节血管壁结构和功能中有重要作用,参与心血管疾病的病理生理过程。e NOS活性的改变除了常见的磷酸化途径外,还涉及乙酰化、谷胱甘肽化、蛋白质间的相互作用等机制,笔者将简要阐述e NOS的基本结构和功能、e NOS活性的调节途径及其在心血管疾病中的研究进展。
【Abstract】 Endothelial nitric oxide synthase(e NOS) is a major enzyme responsible for nitric oxide(NO) production. Both e NOS and NO play an important role in regulating vessel structure and function through participating in the physiological and pathological process. Generally, the phosphorylation of e NOS has been recognized as a common mechanism for regulation of e NOS function. However, recent studies have demonstrated several other new mechanisms, such as acetylation, S-glutathionylation, and protein–protein interaction. This review highlights the basic structure and functions of e NOS as well as the relationship between regulation of e NOS activity and cardiovascular diseases.
