【摘要】 目的探讨Ki-67在胃癌组织中的表达及其临床意义。方法采用免疫组化技术,检测326例胃腺癌组织及相应326例正常胃黏膜组织中Ki-67蛋白的表达情况。采用半定量RT-PCR实验检测,随机选取10例胃腺癌组织和相对应的正常胃黏膜组织中Ki-67mRNA的表达情况,应用χ2检验、Kaplan-Meier生存分析及COX回归分析与患者临床病理特征及预后的关系。结果胃癌组织中Ki-67蛋白和mRNA明显高于正常胃黏膜组织(P<0.05)。Kaplan-Meier生存分析显示Ki-67蛋白表达与胃癌患者的预后密切相关(χ2=8.086,P=0.005),COX回归分析显示Ki-67为胃癌患者预后的独立因素(P=0.002)。结论 Ki-67的表达与胃癌预后密切相关,为胃癌患者预后的独立因素。更多还原

【Abstract】 Objective To investigate the Ki- 67 expression and its clinical significance in gastric cancer. Methods The expressions of Ki- 67 protein in 326 adenocarcinoma of gastric structures and in the corresponding 326 normal gastric mucosa structures will be tested by applying Immunohistochemical Technique. The expressions of Ki- 67 mRNA in 10 random chosen cases of adenocarcinoma of gastric structures and its corresponding normal gastric mucosa structures will be tested by semi- quantitative RT- PCR. The associationof Ki- 67 expression with clinicopathologic features and prognosis in gastric cancer patients were analyzed with various statistical methods,such as χ2- test,Kaplan-Meier method and COX proportional hazard model. Results Ki- 67 protein and Ki- 67 mRNA in the gastric cancer structure clearly is more than that in normal gastric mucosa structure( P < 0. 05). The gastric cancer patient’s survival rate with high expression of Ki- 67 is dramatically lower than that with low expression of Ki- 67. The expression of Ki- 67 is closely related to stomach cancer patient’s prognosis( χ2=8. 086,P = 0. 005). The result of COX proportional hazard model shows that Ki- 67 is the independent prognostic factor of stomach cancer patient( P = 0. 002). Conclusion The expression of Ki- 67 is closely related to the prognosis; Ki- 67 is the independent prognostic factor of gastric cancer.更多还原
