新吉细毛羊KAP24.1基因多态性及羊毛性状的相关性分析
Correlation analysis of the polymorphisms of KAP24.1 gene and the wool traits in Xinji fine-wool sheep
【摘要】 为了解角蛋白关联蛋白(KAP)24.1基因在新吉细毛羊群体中的多态性与其毛用性状的关联性,试验采用DNA测序法对新吉细毛羊基因组编码区进行测序,找出基因突变位点,然后采用PCRSSCP技术对497只新吉细毛羊KAP24.1基因编码区序列进行多态性分析,并利用最小二乘模型对其多态性与产毛量、纤维直径、拉伸长度进行关联性分析。结果表明:在编码区有两处突变点,一处在348 bp处发生C→T的突变,脯氨酸突变为亮氨酸;另一处在414 bp处发生T→C的突变,丝氨酸突变为脯氨酸,均属于错义突变,KAP24.1基因的AA、AB、BB和CC基因型与纤维直径不相关(P>0.05),BB基因型的产毛量和拉伸长度显著大于AB基因型、CC基因型(P<0.05)。说明KAP24.1基因可以作为新吉细毛羊产毛量和拉伸长度主要候选基因之一,BB基因型可以作为分子标记用于预测绵羊产毛量和拉伸长度。
【Abstract】 To understand the correlation between the polymorphisms of KAP24. 1 gene in the population of Xinji fine- wool sheep and the wool traits,DNA sequencing was used to sequence the genomic coding region of Xinji fine- wool sheep to find out gene mutations. And then PCR-SSCP technique was used to analyze the polymorphisms of the coding region sequence of KAP24. 1 gene in 497 Xin Ji fine wool sheep. The correlation between the polymorphisms and production traits including wool yield,wool fiber diameter,and the stretched length was analyzed using a least square linear model. The results showed that there were two mutations in the coding region,thereinto,one mutation from C to T was found at 348 bp,and proline was mutated to leucine; the other mutation from T to C was found at 414 bp,and serine was mutated proline.However,both mutations belonged to the missense mutation. The four kinds of genotypes in the KAP24. 1 gene including AA,AB,BB,and CC were not associated( P > 0. 05) with wool fiber diameter,and the wool yield and the stretched length of BB genotype was significantly greater than( P < 0. 05) that of AB and CC genotypes. The results indicate that the KAP24. 1gene can be used as one of the main candidate genes for the wool yield and the stretched length of Xinji fine- wool sheep,and BB genotype can be used as a molecular marker in predicting sheep wool yield and the stretched length.
