【摘要】 目的检测老年哮喘患者的血清miR-221表达水平,并评价血清miR-221对老年哮喘的诊断价值。方法选取我院109位老年哮喘患者(实验组)和115位健康老年人(对照组)。采用RT-PCR检测2组老年人血清miR-221表达水平,并对结果进行ROC曲线绘制。Elisa法测定2组老年人血清白细胞介素(IL)-4和IL-12浓度,同时对2组老年人进行肺功能检测。最后应用Spearman相关分析计算血清miR-221表达水平与血清细胞因子浓度和肺功能参数的相关性。结果与对照组比较,实验组患者血清miR-221表达水平显著升高,差异有统计学意义(P<0.05)。ROC曲线绘制结果显示,血清miR-221的诊断阈值为1.37。实验组和对照组患者的血清IL-4浓度、血清IL-12浓度、FVC值、FEV1值和PEF值差异均存在统计学意义(P<0.05)。用Spearman相关分析发现,血清miR-221表达水平与血清IL-4浓度、血清IL-12浓度、FVC值、FEV1值和PEF值均存在显著相关性(P<0.05)。结论血清miR-221的表达水平可以作为老年哮喘病情判断指标,同时也为老年哮喘的临床治疗开辟了新思路。更多还原

【Abstract】 Objective To investigate the expression levels of serum miR-221 in senile patients with asthma and evaluate its potential value in the diagnosis of senile asthma. Methods One hundred and nine senile patients with asthma( the experimental group) and 115 healthy senile people( the control group) were recruited for the study. The expression levels of serum miR-221 of the2 groups were detected by RT-PCR,and the receiver operating characteristic( ROC) curve was subsequently obtained. The concentrations of serum IL-4 and IL-12 of the 2 groups were measured by ELISA,and pulmonary function test( PFT) was performed as well. Finally,Spearman method was performed for correlation analysis between serum miR-221 expression levels and serum cytokine concentrations or PFT parameters. Results Compared with that of the healthy senile people,the expression levels of serum miR-221 in the senile patients with asthma were significantly elevated,and statistical significance could be seen,when comparisons were made between them( P < 0. 05). The diagnostic cutoff value of serum miR-221,confirmed by the ROC curve,was 1. 37. There was statistical significance in IL-4 concentrations,IL-12 concentrations,FVC,FEV1 and PEF between the senile patients with asthma and healthy senile people( P < 0. 05). Spearman correlation analysis indicated that there was significant correlation between serum miR-221 expression levels and serum IL-4 concentrations,serum IL-12 concentrations,FVC,FEV1 or PEF( all P < 0. 05). Conclusion The expression level of serum miR-221 could be used as an indicator for the seriousness of senile asthma,and at the same time,it might open up a new avenue for clinical treatment of senile asthma.更多还原
