不完全睡眠剥夺对大鼠甲状腺功能影响的研究
Thyroid function and immune status changes of Wistar rats during partial sleep deprivation
【摘要】 目的研究不完全睡眠剥夺对Wistar大鼠甲状腺功能及相关免疫状态的影响,并分析其可能的机制。方法健康3月龄雄性SPF级Wistar大鼠48只,随机分为睡眠剥夺18 h组(间歇6 h)、睡眠剥夺22 h组(间歇2 h)、持续光照组(24 h持续光照)、对照组(模拟正常12 h拟光照,12 h黑暗的环境),共4组,每组12只大鼠,利用"小平台水环境法"建立大鼠睡眠剥夺模型。中。21 d后检测大鼠血清三碘甲腺原氨酸(TT3)、四碘甲腺原氨酸(TT4)、血清游离三碘甲腺原氨酸(FT3)、游离甲状腺素(FT4)、促甲状腺激素(TSH)、甲状腺球蛋白抗体(Tg抗体)和甲状腺过氧化物酶抗体(TPO抗体)水平。结果睡眠剥夺组大鼠睡眠剥夺实验终末体重较起始体重下降(P<0.01)、睡眠剥夺22 h组的甲状腺相对质量高于对照组(均P<0.01);与对照组相比,睡眠剥夺18 h组血清TT3、TT4、FT4、TSH水平上升(P<0.05,P<0.01);与对照组相比,睡眠剥夺22 h组血清TT3、Tg抗体、TSH水平上升(P<0.05),TT4、FT4水平下降(P<0.01)。结论不完全睡眠剥夺使大鼠甲状腺功能受损,并影响大鼠免疫功能。
【Abstract】 Objective To understand the effects of long-term partial sleep deprivation on function changes of thyroid as well as related immune status and explore their probable mechanisms. Methods Forty eight male Wistar rats aged three months were randomly divided into four groups,12 in each, partial sleep deprivation group 1(SD1 group), partial sleep deprivation group 2(SD2 group), treatment control group(TC group) and cage control group(CC group). The sleep deprivation model was established by flowerpot technique. SD1 group, SD2 group and TC group were all placed in 24-hour proposed lighting environment. The rats in SD1 group were deprived of sleep for 18 hours per day with an intermission for six hours, while rats in SD2 group were deprived of sleep for 22 hours per day with an intermission for two hours. The rats in CC group were placed in12 hours light and 12 hours dark environment. After twenty-one days, the thyroid tissues were collected for weight recording,the blood was collected for determination of total T3(TT3), total T4(TT4), free T3(FT3), free T4(FT4), thyroid stimulating hormone(TSH), anti-thyroglobulin(Tg Ab) and anti-thyroid peroxidase(TPOAb) in serum among different groups. Results The terminal weight of the mice in SD group was significantly lower compared with their initial weight and compared with CC group, the relative mass of thyroid in SD2 group was significantly higher(P<0.01). In SD1 group, compared with CC group, serum TT3 and TT4were obviously higher(P<0.05), serum FT4 and TSH were significantly higher(P<0.01), serum FT3, Tg Ab and TPOAb had a rising trend(P>0.05). In SD2 group, compared with CC group, serum TT3, Tg Ab and TSH obviously higher(P<0.05), serum TT4 and FT4were significantly lower(P<0.01), serum FT3 and TPOAb had a rising trend(P>0.05). Conclusion Long-term partial sleep deprivation may have adverse effect on thyroid function and immune function in rats.
