金纳米颗粒的生物合成及在光热抗肿瘤中的应用
Biosynthesis of gold nanoparticles and application in photothermal treatment of tumor
【摘要】 利用菠菜提取物还原制备金纳米颗粒(AuNPs),通过X射线粉末衍射、紫外-可见吸收光谱、扫描电镜、透射电镜等手段对AuNPs进行了表征。生物合成的AuNPs被提取物中生物活性分子包裹,不仅具有良好的生物相容性和水分散性,而且还可以很好地与肿瘤细胞表面的受体蛋白分子结合,使AuNPs深入肿瘤细胞。用红外热成像研究AuNPs光热效应,还进行细胞存活率测试和细胞荧光成像研究。当AuNPs浓度为60μg/mL时,孵化24h后Hela细胞的存活率为91.2%。说明AuNPs具有很好的生物相容性。同样浓度,在近红外光照10min后,Hela细胞的存活率为64.5%。结果表明,金纳米颗粒表现出显著的光热作用,可用于光热杀死肿瘤细胞。
【Abstract】 Gold nanoparticles(AuNPs)were prepared by bioreduction of chloroauric acid with spinach extracts,and characterized by X-ray powder diffraction,UV-Vis absorption spectra,SEM and TEM.Green synthesis of AuNPs were encapsulated by bioactive molecules in spinach extracts,not only had good biocompatibility and water dispersibility,and can bind well with the receptor protein molecules high expression on tumor cells surface,thus made AuNPs into tumor cells.The photothermal effect of gold nanoparticles under 808 nm NIR radiation were studied using an infrared thermal camera,and the survival assay and fluorescence imaging of tumor cells were investigated as well.When the concentration of AuNPs was 60μg/mL,the survival rate of HeLa cells was 91.2% after incubating for 24 h,which indicated that AuNPs had good biocompatibility.Under the same concentration,the survival rate of Hela cells was 64.5% after the near-infrared laser irradiation for 10 min.The results showed that the gold nanoparticles possessed a remarkable photothermy,and can be used for photothermal treatment of cancer cells.
