四唑染色法快速测定茄子种子生活力的研究
Study on Rapid Determination of Seed Viability of Eggplant by TTC Method
【摘要】 根据四唑(2,3,5-Triphenyl tetrazolium chloride,TTC)染色法测定种子生活力的原理,研究不同浓度四唑溶液、不同染色温度和不同染色时间对茄子(Solanum melongena L.)种子生活力的影响,探索测定茄子种子生活力的最佳条件。结果表明,四唑溶液浓度、染色温度、染色时间三者之间存在一定的相关性,通过改变三者中的一者或两者可以达到改变其他两者或一者的目的,因此,可根据实际需要来确定测定条件。在比较四唑染色法测定茄子种子生活力的不同条件后,认为处理组合11(浸种温度20℃、浸种时间18 h、染色液浓度1.0%、染色温度35℃、染色时间18 h)为最理想的处理组合,是TTC法快速测定茄子种子生活力的适宜方法。
【Abstract】 Based on the principle of tetrazolium(2,3,5-Triphenyl tetrazolium chloride, abbreviation TTC) staining method for determination of seed viability,the effects of concentration of tetrazolium solution, staining temperature and staining time on seed viability of early-maturity eggplant(Solanum melongena L.) were studied to figure out the optimum conditions for determination of eggplant seed viability. The results showed that there was a certain correlation among the concentration of tetrazolium solution, staining temperature and staining time,by changing one or two factors of the three can achieve the purpose of changing the other two or one factor,therefore,it’s possible to determine the optimum conditions according to actual need.After comparing the different combinations of conditions of TTC method for testing early-maturity eggplant seed viability,it was concluded that processing combination 11(soaking seeds in water at 20 ℃ for 18 h,concentration of the tetrazolium solution at1.0%,staining at 35 ℃ for 18 h) is the optimum processing combination, thus suitable for rapid determination of early-maturity eggplant seed viability.
