DNA-PKcs影响肝癌耐药细胞Bel-7402/5-Fu增殖及机制的研究
Study on the effect of DNA-PKcs on the proliferation and the mechanism of the multidrug-resistant hepatocellular carcinoma cell line Bel7402/5-Fu
【摘要】 目的检测DNA-PKcs对肝癌耐药细胞Bel-7402/5-Fu增殖的影响及其相关机制,探讨DNA-PKcs在肝癌多药耐药的发生发展中的作用。方法采用阳离子脂质体法将siDNA-PKcs转染Bel-7402/5-Fu细胞,cck-8法检测细胞增殖情况;流式细胞技术检测细胞周期;Western blot检测DNA-PKcs、TS蛋白的表达情况。结果实验组与对照组比较,细胞增殖减少(P<0.05);实验组S期细胞与对照组比较,细胞数减少(P<0.05);DNA-PKcs、TS蛋白表达结果显示,实验组相比对照组蛋白表达下调,差异有统计学意义(P<0.05)。结论 DNA-PKcs的表达沉默能抑制肝癌耐药细胞Bel-7402/5-Fu的细胞增殖;其机制可能与沉默DNA-PKcs蛋白表达后,TS蛋白表达下调有关。
【Abstract】 Objective To detect the effect of DNA-PKcs on the proliferation of and its mechanism of the multidrug-resistant hepatocellular carcinoma cell line Bel7402/5-Fu,and to investigate the role of DNA-PKcs in the development of multidrug resistance.Methods Transiently was transfected into Bel7402/5-Fu cells via cathodolyte liposome transfection method,and cell proliferation was detected by CCK-8assay.The cell cycle was detected by flow cytometry.Western blot(WB)tested protein expression of DNA-PKcs and TS.Results CCK-8results showed that the experimental group compared with the control group,the cell proliferation was cells in the experimental group was decreased(P<0.05)compared with the control group.WB detection of DNA-PKcs and TS protein expression showed decreased expression of the experimental group than the control group(P<0.05),and there were statistical significantly differences.Conclusion The silence of DNA-PKcs can inhibit decreased(P<0.05).The results of flow cytometry showed that the number of S phase Bel7402/5-Fu cell proliferation,and possible mechanism of silencing DNA-PKcs expression after down-regulation of TS protein expression.
