黄瓜香水提取物对四氯化碳诱导大鼠肝纤维化的影响

【摘要】 探讨黄瓜香对肝纤维化大鼠肝组织α-平滑肌肌动蛋白(α-SMA)及生长转化因子β1(TGF-β1)表达的影响。将48只大鼠随机分为正常对照组、模型组、生理盐水组和黄瓜香干预组,分析仪检测血清ALT、AST、ALP、HA、LN、PCⅢ型胶原,HE染色显示肝的显微结构,SABC法免疫组织学染色显示α-SMA及TGF-β1。结果表明:与模型组和生理盐水组比较,黄瓜香组ALT、AST、ALP、HA、LN、PCⅢ型胶原含量均明显降低(P<0.01);黄瓜香干预组有较正常肝小叶结构,少见纤细的纤维间隔,有炎性细胞少量浸润;模型组、正常对照和黄瓜香干预组α-SMA及TGF-β1表达的平均灰度值比较均有显著性差异(P<0.01)。黄瓜香可明显减少纤维化组织面积,其在治疗肝纤维化方面有开发价值。

【Abstract】 The effects of VDG on the expression of smooth muscle actin alpha( α-SMA) and transforming growth factor-β1( TGF- β1) in rats with hepatic fibrosis were studied. Forty- eight rats were randomly divided into four groups: normal control group,model control group,normal sodium group and interrupt group. The contents of ALT,AST,ALP, HA, LN and PC Ⅲ were measured by full automatic biochemical analyzer and ELISA, hepatic microstructure was showed by HE staining, the expression of α- SMA and TGF- β1was showed by immunohistochemistry dyeing. Compared with the model group,the contents of ALT,AST,ALP,HA、LN and PCⅢremarkably decreased( P<0. 01). In interrupt group,the hepatic lobe structure was normal,the slender textile fiber gap was minority,and there was the few inflammatory cell infiltration. There was significant difference in the average grey degree of α-SMA and TGF-β1among normal control group,model control group and interrupt group( P<0. 01).The alcohol extract from VDG showed the protective effects for liver fibrosis and could alleviate the degree of degeneration and necrosis induced by CCl4.
